仿生提取ICP-MS法测定冬虫夏草8种重金属含量及健康风险评价

Determination of 8 Heavy Metals in Cordyceps Sinensis by Bionic Extraction-ICP-MS and Health Risk Assessment

摘要 目的:建立冬虫夏草中铅(Pb)、镉(Cd)、汞(Hg)、砷(As)、铜(Cu)、银(Ag)、锡(Sn)和锑(Sb)8种重金属的仿生提取含量测定方法,并对其健康风险进行评价。方法:采用人工胃液、人工肠液仿生提取冬虫夏草中可溶性重金属,ICP-MS法测定重金属含量,靶标危害系数(THQ)法评价健康风险。结果:8种重金属在各自质量浓度范围内线性关系良好(R2> 0.997 0),平均回收率84.05%~114.71%(RSD 2.9%~4.9%)。冬虫夏草中As、Hg、Cu、Cd、Sb、Pb、Ag总量均值依次为12.122、0.644、15.828、0.095、0.027、0.636、0.075 mg·kg-1,未检出Sn。As、Hg、Cu、Cd、Pb、Ag在人工胃液溶出率大于人工肠液,其在人工胃液中的溶出率均值为19.69%~52.62%,在人工肠液中的溶出率均值为0%~16.05%;Sb在人工肠液的溶出率25.54%大于人工胃液的溶出率14.68%。健康风险评价结果显示,As、Hg、Cu、Cd、Pb、Ag、Sb的THQ均<1,表明冬虫夏草中上述8种重金属不会对人体造成明显的健康危害。结论:建立的方法简便、准确、可靠,适用于冬虫夏草中上述8种重金属的测定和健康风险评价,仿生提取测定冬虫夏草8种重金属的含量数据及基于该数据的健康评价结果可为冬虫夏草重金属安全性深入研究提供参考。 Objective:A bionic extraction method to determinate the enrichment of corresponding health risk of eight heavy metals(Pb,Cd,Hg,As,Cu,Ag,Sn,and Sb)in Cordyceps sinensis was established was evaluated.Methods:The soluble heavy metals in C.sinensis were extracted by artificial gastric juice and artificial intestinal juice and the enrichement of the heavy mentals was further determined by ICP-MS.Target hazard coefficient(THQ)was employed to evaluated the health risk.Results:Good linear relationships of eight heavy metals was calculated in mass concentration ranges(R2>0.9970),respectively.Averaged recoveries were 84.05%-114.71%(RSD 2.9%-4.9%).The averaged contents of As,Hg,Cu,Cd,Sb,Pb,and Ag in C.sinensis were 12.122,0.644,15.828,0.095,0.027,0.636,and 0.075 mg·kg-1,respectively,in which Sn was not detected.The dissolution rate of As,Hg,Cu,Cd,Pb,and Ag in artificial gastric juice emerged to be higher than in artificial intestinal juice.The averaged dissolution rate of As,Hg,Cu,Cd,Pb,and Ag in artificial gastric juice was 19.69%-52.62%,The averagde dissolution rate in artificial intestinal juice was calculated asthe range of 0%-16.05%.The dissolution rate of Sb in artificial intestinal fluid was 25.54%,which was higher than that in artificial gastric fluid(14.68%).The results of health risk assessment indicated that the THQ of As,Hg,Cu,Cd,Pb,Ag,and Sb were all less than 1,indicating that the aforementioned eight heavy metals in C.sinensis showed rare posibility to cause obvious health hazards to human body.Conclusion:The established method was simple,accurate,and reliable,the acquired data indicated the established method was apporpriate for the determination and health risk assessment of the aforementioned 8 heavy metals in C.sinensis.The enrichment data of 8 heavy metals in C.sinensis and the healthy evaluation results based on the data might provide quantified date for the in-depth study on safty assessment of heavy metals in C.sinensis.
