健脾祛痰方对限制活动ApoE^(-/-)小鼠体成分/行为学变化的影响

摘要 目的:观察健脾祛痰方对限制活动载脂蛋白E基因敲除(Apolipoprotein E Knockout,ApoE^(-/-))小鼠体成分/行为学变化的影响。方法:将30只ApoE^(-/-)小鼠随机分为模型组,阿托伐他汀组以及健脾祛痰方低浓度组、健脾祛痰方中浓度组、健脾祛痰方高浓度组;将6只C57BL/6J小鼠作为正常组。正常组以普通饲料喂养,其余5组以高脂饲料喂养,阿托伐他汀组进行阿托伐他汀2.4 mg/(kg·d)灌胃,健脾祛痰方低浓度组、健脾祛痰方中浓度组、健脾祛痰方高浓度组以健脾祛痰方[2.97 g/(kg·d)、5.94 g/(kg·d)、11.88 g/(kg·d)]灌胃,正常组和模型组以等体积生理盐水灌胃,12周后进行脾虚痰浊模型评价。实验期间每周观察小鼠的皮毛色泽、活动能力及大便性状,检测体成分,以旷场实验、跑台实验进行动物行为学评估。结果:与正常组比较,模型组小鼠旷场实验运动总距离减少,跑台实验跑步力竭时间降低,体脂降低,差异有统计学意义(P <0.01)。与模型组比较,健脾祛痰方低浓度组、健脾祛痰方中浓度组、健脾祛痰方高浓度组小鼠旷场实验运动总距离呈上升趋势,但差异无统计学意义(P> 0.05);健脾祛痰方中、高浓度组小鼠跑台实验跑步时间增加,差异有统计学意义(P <0.05,P <0.01);阿托伐他汀组和健脾祛痰方低浓度组跑步时间均呈上升趋势,但差异无统计学意义(P> 0.05);健脾祛痰方低浓度组、健脾祛痰方中浓度组、健脾祛痰方高浓度组小鼠体脂呈上升趋势,但差异无统计学意义(P> 0.05);阿托伐他汀组小鼠体脂显著增加,差异有统计学意义(P <0.01)。结论:健脾祛痰方可增加限制活动ApoE^(-/-)小鼠的行为活动,但对体成分改善作用不显著。 Objective:To observe the effect of the Jianpi Qutan prescription(健脾祛痰方) on the changes of body composition/behavior in ApoE^(-/-)mice with restricted activities.Methods:A total of 30 ApoE^(-/-)mice were randomly divided into the model group,the Atto vastatin group,the low concentration group,the middle concentration group and the high concentration group of the Jianpi Qutan prescription,and 6 C57BL/6J mice were taken as the normal group.The normal group was fed with normal diet and the other 5 groups were fed with high fat diet.The Atto vastatin group was fed with Atto vastatin 2.4 mg/(kg ·d).The low concentration group,the middle concentration group and the high concentration group were fed with the Jianpi Qutan prescription [2.97 g/(kg·d),5.94 g/(kg·d),11.88 g/(kg·d)],and the normal group and the model group were fed with the same volume of normal saline.The model of spleen deficiency and phlegm turbidity was evaluated after 12 weeks.During the experiment,the fur color,activity ability,stool characteristics and body composition of mice were observed and recorded every week,and the open field test and treadmill test were used to evaluate the animal behavior.Results:Compared with the normal group,the total exercise distance of mice in the model group was reduced in the open field test,the exhaustion time of mice in the treadmill test and the body fat of mice in the model group were decreased,with statistical significance(P<0.01).Compared with the model group,the total exercise distance of mice in the low concentration group,the middle concentration group and the high concentration group of the Jianpi Qutan prescription in open field test showed an increasing trend,but the difference was not statistically significant(P>0.05).The running time of mice in the middle concentration group and the high concentration group of the Jianpi Qutan prescription was increased in treadmill test,and the difference was statistically significant(P<0.05,P<0.01).The running time of the Atto vastatin group and the low concentration group of the Jianpi Qutan prescription showed an increasing trend,but there was no statistical significance(P>0.05).Body composition test of the Jianpi Qutan prescription showed an increasing trend of body fat in the low concentration group,the middle concentration group and the high concentration group of the Jianpi Qutan prescription,but there was no statistical significance(P>0.05).The body fat in the Atto vastatin group was significantly increased,and the difference was statistically significant(P<0.01).Conclusion:The Jianpi Qutan prescription can increase the behavioral activity of ApoE^(-/-)mice with restricted activity,but has no significant effect on body composition.
