基于网络药理学分析青蒿鳖甲汤治疗癌性发热的作用机制

摘要 目的 采用中药网络药理学初步分析青蒿鳖甲汤在治疗癌性发热方面的作用与机制。方法 借助TCMSP及HERB数据库初步筛查出青蒿鳖甲汤相关优效活性成分,利用Swiss Target Prediction和PubChem平台预测优效活性成分的相关作用靶点并在GeneCards上筛选治疗癌性发热的相关作用靶标,后在韦恩图上进行映射。通过STRING数据库和Cytoscape 3.9.1软件构建PPI网络以获取青蒿鳖甲汤在治疗癌性发热方面的关键靶点,最后借助DAVID数据库对青蒿鳖甲汤抗癌性发热作用的关键靶点进行GO和KEGG富集分析。结果 得到青蒿鳖甲汤药物成分与癌性发热疾病共132个共同靶点,KEGG通路125条。结论 本研究初步分析出青蒿鳖甲汤中含有槲皮苷、山奈酚、木犀草素、淫羊藿素等多种成分,这些成分可能作用于VEGFA、CASP3、AKT1、MMP9、TNF、HIF1A、EGF、TP53、PTGS2、IL6等治疗靶点上,并通过调控TNF、IL-17、PI3K-Akt等信号通路发挥治疗癌性发热的作用,为临床运用青蒿鳖甲汤提供了理论参考和依据。 Objective Using traditional Chinese medicine network pharmacology to conduct initial analysis on the role and mechanism of Qinghao Biejia decoction in treating cancerous fever.Methods Utilizing the TCMSP and HERB databases for initial screening of active ingredients related to Qinghao Biejia decoction.Predicting relevant target proteins of these active ingredients using Swiss Target Prediction and PubChem platforms,filtering target spots associated with "treating cancerous fever" on GeneCards,followed by mapping on Venn diagrams.Constructing a PPI network through STRING database and Cytoscape 3.9.1 software to identify key target spots for the treatment of cancerous fever using Qinghao Biejia decoction.Finally,conducting GO and KEGG enrichment analyses on key target spots associated with the anticancer fever effects of Qinghao Biejia decoction with the assistance of the DAVID database.Results A total of 132 common target spots were identified between the medicinal ingredients of Qinghao Biejia decoction and cancerous fever disease,involving 125 KEGG pathways.Conclusion This study has conducted a preliminary analysis revealing that Qinghao Biejia decoction contains various components such as quercetin,kaempferol,luteolin,icariin,and others.These components potentially target spots therapeutic points including VEGFA,CASP3,AKT1,MMP9,TNF,HIF1A,EGF,TP53,PTGS2,IL6 and may exert their effects in treating cancerous fever by modulating signaling pathways such as TNF,IL-17,PI3K-Akt.This offers theoretical references and foundations for the clinical application of Qinghao Biejia decoction.
