HPLC法测定利咽含漱液中木犀草苷含量及^(60)Co-γ射线辐照对其含量的影响

Determination of galuteolin in Liyan gargle by HPLC and the effect of 60Co-γradiation on its content

建立了一种采用HPLC法测定利咽含漱液中木犀草苷含量的方法,并考察不同剂量^(60)Co-γ射线辐照对其含量的影响.用Agilent ZORBAX Eclipse XDB-C18(4.6×250 mm,5μm)色谱柱,以乙腈-0.1%磷酸(20∶80)为流动相,流速为1.0 mL·min^(-1),柱温为30℃;检测波长为350 nm,进样量为10μL,用外标法计算含量.研究表明,木犀草苷含量测定方法的专属性良好;浓度在1.8954~47.3858μg·mL^(-1)范围内与峰面积呈良好的线性关系,线性方程为Y=31.13X-15.793,相关系数r=0.9997(n=6);平均加样回收率为99.73%(n=6);精密度的RSD值为0.58%(n=6),稳定性的RSD值为1.26%(n=6),重复性的RSD值为0.87%(n=6),耐用性的RSD均小于3%.经^(60)Co-γ射线辐照后利咽含漱液中木犀草苷含量下降,下降幅度与辐照剂量呈正相关.本方法专属性强、准确度高,适用于利咽含漱液中木犀草苷含量测定.利咽含漱液不宜进行^(60)Co-γ射线辐照灭菌.

An HPLC method was established to determine the content of galuteolin in Liyan gargle,and the effect of different do-ses of^(60)Co-γradiation was investigated on the content of galuteolin in Liyan gargle.Using Agilent ZORBAX Eclipse XDB-C18(4.6×250 mm,5μm)as chromatographic column,with acetonitrile-0.1%phosphoric acid(20:80)as the mobile phase,flow rate of 1.0 mL·min^(-1),and column temperature of 30℃;the detection wavelength was 350 nm,and the injection volume was 10μL,the content was calculated using the external standard method.Research showed that the specificity of the method for determining the content of galuteolin was good;there was a good linear relationship between the peak area and the range from 1.8954~47.3858μg·mL^(-1),with a linear equation of Y=31.13X-15.793 and a correlation coefficient of r=0.9997(n=6);the average sample recovery rate was 99.73%(n=6);the RSD value for precision was 0.58%(n=6),the RSD value for stability was 1.26%(n=6),the RSD value for repeatability was 0.87%(n=6),and the RSD for durability was less than 3%.After^(60)Co-γradiation,the content of galuteolin in Liyan gargle decreased,and the decrease was positively correlated with the ra-diation dose.This method had strong specificity and high accuracy,and was suitable for the determination of galuteolin content in Liyan gargle.Liyan gargle should not be used for^(60)Co-γradiation sterilization.
