鱼鳔氨基酸组成分析及抗氧化活性研究

摘要 以鱼鳔为研究对象,采用氨基酸自动分析仪器对鱼鳔蛋白及其多肽的氨基酸种类和含量进行测定并分析。结果表明:鱼鳔蛋白及其多肽均含有16种氨基酸,鱼鳔蛋白总氨基酸含量为92043.85 ng/mg,鱼鳔胰蛋白酶酶解多肽总氨基酸含量为79402.03 ng/mg,鱼鳔碱性蛋白酶酶解多肽总氨基酸含量为71978.95 ng/mg,鱼鳔胃蛋白酶酶解多肽总氨基酸含量为335053.03 ng/mg。随后以鱼鳔蛋白为研究对象,对鱼鳔蛋白进行体外和体内抗氧化实验研究。鱼鳔蛋白对NO、羟基自由基有清除能力,对亚铁离子以及铜离子有螯合能力;体内对氧化损伤线虫的存活率的影响在一定范围内均随蛋白浓度的增加而增强,表现出较好的抗氧化能力。 One of the material bases of swim bladder is the composition of amino acids and amino acid components.The amino acid types and contents of swim bladder protein and its polypeptides were determined and analyzed by an automatic amino acid analyzer.The results showed that both the swim bladder protein and its polypeptide contained 16 amino acids.The total amino acid content of fish bladder protein,fish bladder trypsin hydrolyzed peptide,fish bladder alkaline protease hydrolyzed peptide,and fish bladder pepsin hydrolyzed peptide was 92043.85 ng/mg,79402.03 ng/mg,71978.95 ng/mg,335053.03 ng/mg.Subsequently,fish bladder protein was used as the research object to conduct in vitro antioxidant experiments,release performance experiments,and in vivo experimental studies on the impact of oxidative damage on the survival rate of nematodes.The experimental results showed that fish swim bladder peptides has the ability to scavenge NO and hydroxyl radicals,and chelate ferrous and copper ions;The impact of oxidative damage on the survival rate of nematodes in vivo increases with the increase of protein concentration within a certain range,demonstrating good antioxidant ability.
