1800MHz电磁辐射暴露对大鼠外周血单个核细胞代谢组的影响

摘要 目的 分析电磁辐射暴露对大鼠外周血单个核细胞(PBMCs)代谢物变化的影响。方法 将24只SD大鼠随机分成电磁辐射暴露组(EMR组)和假暴露组(Sham组)。EMR组大鼠置于中心频率1 800 MHz、平均功率密度284.6 V/m的电磁混响箱内,每天暴露10 min,连续暴露7 d;Sham组辐照功率为0 W,其他条件及操作与EMR组相同。7 d后,收集大鼠外周血并分离PBMCs进行液相色谱/质谱联用(LC-MS/MS)非靶向代谢组学分析。使用偏最小二乘法-判别分析(PLS-DA)进行主成分分析,单变量分析筛选差异代谢物,再联合mzCloud和ChemSpider数据库对检测到的代谢物进行鉴定,构建代谢通路以分析电磁辐射暴露对大鼠PBMCs代谢组的影响。结果 Sham组与EMR组SD大鼠PBMCs正负离子模式下共检测到差异代谢物2 017种,正离子模式下1 611种,其中847种上调,764种下调;负离子模式下406种,其中220种上调,186种下调。正负离子模式下检测到的差异代谢物包括氨基酸、肽类、有机酸、胆汁酸等。KEGG富集分析显示,正负离子模式下差异代谢物涉及的代谢通路共64个,包括氨基酸代谢,脂类代谢,细胞生长、增殖及死亡,氧化应激,炎症反应等。结论 模拟职业环境电磁辐射暴露可以对SD大鼠PBMCs代谢组产生明显影响,其差异代谢产物有望成为电磁辐射暴露的生物标志物。 Objective By simulating the relevant anatomical structure of the lateral fissure-insular lobe through the cadaver head specimen,it can provide reference for clinicians to carry out insular gliomas resection,thereby improving the total tumor resection rate while maximizing the protection of brain tissue function.Methods Anatomical study of lateral fissure,insular lobe,and middle cerebral artery regions on 10 cadaveric head specimens were conducted.The relationship among the important structures were observed by taking photos,and the relevant parameters were measured.Results The central sulcus of the island was parallel to the central sulcus on the surface of the brain,which divides the insular lobe into the front half and the back half.The front half consists of 3 to 5 short insular gyri,the back half consists of 2 long insular gyri.The front half of insular lobe was covered by the trigone of the inferior frontal gyrus,the tectum and the anterior central gyrus.The upper part of the back half of the insular lobe was covered by the posterior central gyrus,the supramarginal gyrus,
