蒙药消肿九味散对痛风性关节炎大鼠尿酸及炎性细胞因子的影响

Effect of Mongolian medicine Xiaozhong Jiuwei Powder on uric acid and inflammatory cytokines in rat with gouty arthritis

目的观察蒙药消肿九味散外用对痛风性关节炎大鼠尿酸及炎性细胞因子的影响。方法将40只雄性Wistar大鼠随机分为正常组、模型组、扶他林乳胶剂组、云南白药喷剂组和蒙药消肿九味散组与,每组8只。除正常组外,其余组大鼠采用氧嗪酸钾、尿酸钠制备痛风性关节炎模型。造模成功后,扶他林乳胶剂组给予扶他林乳胶剂外敷,云南白药喷剂组给予云南白药喷雾剂外用,蒙药消肿九味散组给予蒙药消肿九味散(与鸡蛋清以1∶2的比例混合调制成糊状)外敷,均1次/d,干预14 d。检测大鼠血清尿酸及前列腺素E 2(PGE 2)、白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)水平,HE染色观察踝关节组织病变情况。结果给药14 d后,各用药组大鼠血清尿酸及PGE 2、IL-1、IL-6、IL-8、TNF-α水平均明显低于模型组(P均<0.05);扶他林乳胶剂组血清尿酸水平均明显低于云南白药喷剂组和蒙药消肿九味散组(P均<0.05),云南白药喷剂组和蒙药消肿九味散组比较差异无统计学意义(P>0.05);各用药组大鼠血清PGE 2水平比较差异均无统计学意义(P均>0.05);扶他林乳胶剂组大鼠血清IL-1、IL-8、TNF-α水平和云南白药喷剂组大鼠血清IL-6、TNF-α水平均明显低于蒙药消肿九味散组(P均<0.05);蒙药消肿九味散组血清IL-8水平明显低于云南白药喷剂组(P<0.05),血清IL-1水平与云南白药喷剂组比较、血清IL-6水平与扶他林乳胶剂组比较差异均无统计学意义(P均>0.05)。相比于模型组,各用药组大鼠踝关节整体结构轻度异常,滑膜炎症程度均有不同改善,炎性细胞浸润及滑膜增生明显减轻,以蒙药消肿九味散组改善最为明显。结论蒙药消肿九味散外用能有效降低痛风性关节炎大鼠血清尿酸和相关炎性细胞因子水平,从而减轻炎性损伤。

Objective It is to observe the effect of external application of Mongolian medicine Xiaozhong Jiuwei Powder on uric acid and inflammatory cytokines in rats with gouty arthritis(GA).Methods Forty male Wister rats were randomly divided into normal group,model group,Voltaren emulsion group,Yunnan Baiyao spray group,Mongolian medicine Xiaozhong Jiuwei Powder group,with 8 rats in each group.The rats of all groups except for the normal group were given potassium oxonate and sodium urate to prepare models of GA.After successful modeling,the Voltaren emulsion group was given external application of Voltaren emulsion,the Yunnan Baiyao spray group was given external application of Yunnan Baiyao spray,and the Mongolian medicine Xiaozhong Jiuwei Powder group was given external application of Mongolian medicine Xiaozhong Jiuwei Powder(mixed with egg white in the ratio of 1∶2 to form a paste),all once daily,continuously treated for 14 days.The serum levels of uric acid(UA),tumor necrosis factor-α(TNF-α),prostaglandin E 2(PGE 2),interleukin-1(IL-1),interleukin-6(IL-6)and interleukin-8(IL-8)in the rats were determined,and the ankle joint tissue lesions were observed by HE staining.Results After 14 days of treatment,the serum levels of UA,PGE 2,IL-1,IL-6,IL-8,TNF-α in the rats of each treatment group were significantly lower than those of the model group(all P<0.05);the serum level of UA of the Voltaren emulsion group was significantly lower than those of the Yunnan Baiyao spray group and the Mongolian medicine Xiaozhong Jiuwei Powder group(all P<0.05),there was no statistically significant difference between the Yunnan Baiyao spray group and the Mongolian medicine Xiaozhong Jiuwei Powder group(P>0.05);the differences in the serum levels of PGE 2 in the rats of each treatment group were not statistically significant(all P>0.05);the serum levels of IL-1,IL-8 and TNF-α in the rats of the Voltaren emulsion group and the serum levels of IL-6 and TNF-α of the Yunnan Baiyao spray group were significantly lower than those of the Mongolian medicine Xiaozhong Jiuwei Powder group(all P<0.05);the serum level of IL-8 in the Mongolian medicine Xiaozhong Jiuwei Powder group was significantly lower than that in the Yunnan Baiyao spray group(P<0.05),and there was no statistically significant differences in the serum level of IL-1 compared with the Yunnan Baiyao spray group,and no statistically significant differences in the serum level of IL-6 compared with the Voltaren emulsion group(P>0.05).Compared with the model group,the overall structure of the ankle joint of rats in each treatment group was mildly abnormal,and the synovial inflammation was improved more or less,and the inflammatory cell infiltration and synovial hyperplasia were significantly alleviated,the improvement was the most obvious in the Mongolian medicine Xiaozhong Jiuwei Powder group.Conclusion External application of Mongolian medicine Xiaozhong Jiuwei Powder can effectively reduce the serum levels of UA and inflammatory cytokines,thus to alleviate inflammation injury in rats with gouty arthritis.
