摘要
目的:研究茴香提取液对四氯化碳(CCl4)复合酒精诱导肝纤维化小鼠的干预作用。方法:取SPF雄性小鼠采用CCl4复合酒精构建小鼠肝纤维化模型,造模成功后随机分为模型组、茴香低剂量组、茴香中剂量组、茴香高剂量组,每组8只,另取8只小鼠作为正常对照组。相应药物干预5周后,全自动生化分析仪检测外周血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平;ELISA法测定透明质酸(HA)、层黏连蛋白(LN);生化法检测肝组织超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;肝组织HE染色和Masson染色观察肝脏组织形态学改变和胶原纤维染色。结果:与正常对照组比较,模型组ALT、AST、HA、LN和MDA水平显著升高(P<0.01),SOD活力显著降低(P<0.01)。HE染色显示,模型组肝细胞排列紊乱,纤维结缔组织增生,大量炎症细胞浸润,可见假小叶形成;Masson染色显示,模型组大量胶原纤维增生,且增生的胶原纤维形成纤维间隔。与模型组比较,茴香各治疗组ALT、AST酶活性及HA、LN水平均显著降低(P<0.05或P<0.01),茴香中、高剂量组MDA水平均显著降低(P<0.05),茴香高剂量组肝组织SOD活力显著增高(P<0.05)。HE染色、Masson染色显示茴香低、中、高剂量组对纤维化程度均有不同改善,以茴香高剂量组改善作用显著。结论:茴香提取液可以保护肝细胞和缓解肝纤维化,减轻CCl4复合酒精诱导小鼠肝纤维化程度。
Objective:To study the effects of fennel extract on hepatic fibrosis induced by carbon tetrachloride(CCl4)combined with alcohol in mice.Methods:SPF male mice were randomly divided into model group,fennel low-dose group,fennel medium-dose group and fennel high-dose group after successful modeling by using CCl4 combined with alcohol,eight mice in each group.Another eight mice were selected as control group.After five weeks of drug intervention,the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in peripheral blood were detected by automatic biochemical analyzer.ELISA was used to detect hyaluronic acid(HA)and laminin(LN).The activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in liver tissues were detected by biochemical method.Liver histomorphological changes and collagen fiber staining were observed by liver tissue HE staining and Masson staining.Results:Compared with control group,the levels of ALT,AST,HA,LN and MDA were significantly increased in model group(P<0.01),while SOD activity was significantly decreased(P<0.01).HE staining showed disordered arrangement of liver cells,fibrous connective tissue hyperplasia,infiltration of a large number of inflammatory cells,and pseudolobular formation in model group;Masson staining showed that there were a large number of collagen fibers proliferate and hyperplastic collagen fibers form fibrous intervals in model group.Compared with model group,the activity of ALT and AST enzymes and the levels of HA and LN significantly decreased in fennel treatment groups(P<0.05 or P<0.01);The levels of MDA decreased in fennel medium-dose and high-dose groups(P<0.05),while the SOD activity in liver tissues significantly increased in fennel high-dosel group(P<0.05).
