IL-2/抗IL-2抗体通过增加中枢神经系统内调节性T细胞数量减轻小鼠颅脑创伤

摘要 目的探讨IL-2/抗IL-2抗体增加颅脑创伤(TBI)小鼠中枢神经系统内调节性T细胞(Treg)数量对小鼠TBI的影响。方法所用动物为10周龄C57BL/6健康雄性小鼠,分为Sham组、TBI组、TBI+IL-2/抗IL-2抗体组。TBI+IL-2/抗IL-2抗体组在造模2 h后连续3 d给予IL-2(1.0μg)+抗IL-2抗体(5.0μg)腹腔注射。提取三组小鼠在3、5、7 d的脑组织进行流式细胞术,检测Treg数量,同时将第7日小鼠脑组织进行免疫荧光染色,以明确TBI后中枢神经系统存在Treg浸润,另外对三组小鼠1、3、5、7 d的行为学结果分别进行分析。同时检测造模第7日各组小鼠脑组织含水量、神经元凋亡及炎症因子表达情况。结果与Sham组相比,TBI组及TBI+IL-2/抗IL-2抗体组小鼠中枢神经系统内确实存在Treg浸润情况,且Treg在脑创伤后逐渐增多,Sham组未见Treg浸润,TBI+IL-2/抗IL-2抗体组相较于TBI组Treg明显增多(P<0.05),包括神经元凋亡减少、脑水肿减轻及炎症因子表达降低,预后明显改善。结论IL-2/抗IL-2抗体可以增加中枢神经系统内Treg的数量并在一定程度上能够减轻小鼠TBI症状。 Objective To investigate the effect of IL-2/anti-IL-2 antibody on traumatic brain injury(TBI)in mice by increasing the number of regulatory T cells(Treg)in the central nervous system.Methods Healthy male C57BL/6 mice aged 10 weeks were divided into Sham group,TBI group and TBI+IL-2/anti-IL-2 antibody group.The TBI+IL-2/anti-IL-2 antibody group was intraperitoneally injected with IL-2(1.0μg)and anti-IL-2 antibody(5.0μg)for 3 consecutive days at 2 h after modeling.Flow cytometry was performed on the brain tissues of the three groups of mice at 3,5 and 7 d to detect the number of Treg.Immunofluorescence staining was performed on the brain tissues of mice on the 7th day to determine the presence of Treg infiltration in the central nervous system after TBI.In addition,the behavioral results of the three groups of mice at 1,3,5,and 7 d were analyzed,respectively.At the same time,the water content of brain tissues,neuronal apoptosis and the expression of inflammatory factors were detected on the 7th day of modeling.Results Compared with Sham group,there was Treg infiltration in the central nervous system of mice in TBI group and TBI+IL-2/anti-IL-2 antibody group,and the number of Treg gradually increased after TBI.There was no Treg infiltration in the Sham group.Compared with TBI group,TBI+IL-2/anti-IL-2 antibody group had a significant increase in Treg(P<0.05),including a reduction in neuronal apoptosis,brain edema,and expression of inflammatory factors,and a significant improvement in prognosis.Conclusion IL-2/anti-IL-2 antibody can increase the number of Treg in the central nervous system and alleviate TBI symptoms in mice to a certain extent.
