大孔树脂富集大花红景天总黄酮及抗氧化活性研究

The Enrichment of Total Flavonoids from Rhodiola crenulata by Macroporous Resin and Evaluation of Antioxidant Activity

摘要 本研究旨在建立一种大花红景天中总黄酮的含量测定方法,运用大孔吸附树脂富集大花红景天中总黄酮,从5种大孔树脂中筛选出最优树脂,并对其纯化工艺进行初步优化,对富集得到的红景天总黄酮的抗氧化活性进行测定。以芦丁作为对照品,采用亚硝酸钠-硝酸铝显色反应比色法,测定大花红景天中的总黄酮含量。运用静态吸附-解吸附、动态吸附-解吸附方法筛选出最优大孔树脂,对洗脱剂浓度、用量等工艺进行优化,并测定大花红景天中总黄酮的抗氧化活性。西藏产大花红景天中总黄酮含量为12.91%,青海产红景天中总黄酮含量为9.85%,D101型大孔树脂为富集大花红景天中总黄酮的最优树脂,最佳洗脱剂浓度为80%乙醇溶液,最佳洗脱剂用量为5 BV。经D101型大孔树脂纯化后,西藏产大花红景天中总黄酮含量达到24.64%,其对DPPH自由基的清除率可达到95.51%。D101型大孔树脂对富集大花红景天中总黄酮具有较好的效果,大花红景天中总黄酮具有较好的抗氧化活性,该研究可为丰富大花红景天的质量评价标准和加快对大花红景天中黄酮类组分的开发利用提供参考。 To establish a method for determining the content of total flavonoids in Rhodiola crenulata.Macroporous adsorption resin was used to enrich the total flavonoids in Rhodiola crenulata.The best resin was selected from 5 types of macroporous resins,and the purification process was preliminarily optimized.Determine the antioxidant activity of the enriched total flavonoids in Rhodiola crenulata.Using rutin as a reference substance,the colorimetric method of sodium nitrite-aluminum nitrate reaction was used to determine the total flavonoids content in Rhodiola crenulata.The best macroporous resin was selected using static adsorption-desorption and dynamic adsorption-desorption methods,and the concentration and dosage of eluent were optimized,and the antioxidant activity of total flavonoids in Rhodiola crenulata was determined.The total flavonoids content in Rhodiola crenulata from Tibet is 12.91%,while that in Rhodiola crenulata from Qinghai is 9.85%.D101 macroporous resin was the best resin for enriching total flavonoids in Rhodiola crenulata.The best eluent concentration is 80%ethanol solution,and the best eluent dosage is 5 BV.After purification with D101 macroporous resin,the total flavonoids content in Rhodiola crenulata from Tibet reached 24.64%.Its clearance rate for DPPH free radicals can reach 95.51%.D101 macroporous resin has a good effect on enriching the total flavonoids in Rhodiola crenulata,and the total flavonoids in Rhodiola crenulata have a good antioxidant activity.This study can provide reference for enriching the quality evaluation standards of Rhodiola crenulata and accelerating the development and utilization of flavonoids in Rhodiola crenulata.
