干冰用于鸡超低温冷冻精液短期保存和运输的可靠性评估

摘要 为解决液氮保存下鸡冷冻精液的异地运输难题,研究对北京油鸡冷冻精液在干冰(-79℃)中进行短期保存,分别保存0 h(对照组)、12 h、24 h、48 h和72 h,以模拟冷冻精液在干冰中的短期运输,并测定干冰不同保存时间的鸡冷冻精液精子质量和受精能力。结果显示:干冰保存后北京油鸡冷冻精液的精子活力、存活率、顶体完整率、质膜完整率和受精率均显著降低(P<0.05),精子畸形率显著增高(P<0.05),且随着干冰保存时间的增加,精子质量和受精能力逐渐下降。研究表明,干冰作为冷源对鸡冷冻精液的精子质量有较大的损害作用,使用干冰难以实现对鸡冷冻精液的有效短期保存和运输。 To address the challenges associated with long-distance transportation of chicken frozen semen under liquid ni⁃trogen storage,this study was conducted to simulate short-term transportation of chicken frozen semen in dry ice(-79℃).Frozen semen samples of Beijing You chickens were stored in dry ice for 0 h(control group),12 h,24 h,48 h and 72 h.And the sperm quality and fertilization ability of the frozen semen samples were then determined.The results showed that the sperm viability,sur⁃vival rate,acrosome integrity rate,plasma membrane integrity rate and fertilization rate of frozen sperm of Beijing You chicken af⁃ter dry ice storage were significantly reduced(P<0.05),the sperm deformity rate was significantly increased(P<0.05),and the sperm quality gradually decreased with the increasing storage time in dry ice.This study demonstrated that dry ice as a cold source significantly impacted quality of chicken frozen sperm,which posed challenges in achieving effective short-term storage and transportation of this biological material.
