甜菜碱缓解皮质酮诱导的产蛋鸡肝脏炎症反应研究

摘要 试验旨在研究饲粮添加甜菜碱缓解皮质酮诱导的产蛋鸡肝脏炎症反应。试验采用单因子完全随机试验设计,将72只130日龄产蛋鸡随机分成3组,分别为对照组、皮质酮组和甜菜碱缓解组,每组8个重复。试验期35 d。对照组和皮质酮组鸡全程饲喂不添加甜菜碱的饲粮,而甜菜碱缓解组鸡全程饲喂添加0.10%甜菜碱的饲粮,试验至28 d时,皮质酮与甜菜碱缓解组鸡均按4 mg/kg体重的剂量注射皮质酮,而对照组鸡注射等体积的酒精生理盐水(含15%酒精),连续注射7 d。结果显示:饲粮添加甜菜碱有效缓解皮质酮诱导的血浆中谷草转氨酶和谷丙转氨酶活性的升高以及肝脏中IL-1βmRNA表达水平的升高(P<0.05);甜菜碱显著缓解皮质酮诱导的IL-1βDNA启动子区甲基化水平的降低(P<0.05)。研究表明,甜菜碱通过DNA甲基化修饰抑制肝脏中IL-1βmRNA表达,从而缓解皮质酮诱导的产蛋鸡肝脏炎症反应。 This experiment was conducted to investigate the alleviating effect of dietary betaine on corticosterone(CORT)-induced hepatic inflammation in laying hens.A completely randomized experimental design was adopted,a total of 72130-dayage laying hens were divided into three groups:the control group,corticosterone group and betaine group.The experiment lasted for 35 days.Chickens in the control and corticosterone groups were fed diet without added betaine for 35 days,while chickens in the betaine group were fed diet supplemented with 0.10%betaine.On day 28 of the experiment,chickens in the corticosterone and betaine groups were daily injected subcutaneously with CORT(4.0 mg/kg body mass),while the chickens in the control group were injected with equal volume solvent(15%ethanol)for 7 days.The results showed that betaine could significantly alleviate CORT-induced up-regulation of plasma glutamic pyruvic transaminase and glutamic oxaloacetic transaminase activities,and he⁃patic IL-1βmRNA expression(P<0.05).Furthermore,dietary betaine could significantly alleviate CORT-induced hypomethyl⁃ation on IL-1βpromoter(P<0.05).It's indicated that dietary betaine could protect laying hens from CORT-induced hepatic in⁃flammation via epigenetic modulation of IL-1βgene.
