摘要
目的探讨驴食草酚(vestitol)对过氧化氢(H_(2)O_(2))引起人脐静脉内皮细胞(HUVEC)损伤的保护作用。方法体外培养HUVEC,通过设置700~900μmol/L H_(2)O_(2)系列浓度筛选H_(2)O_(2)损伤HUVEC浓度。设置1.25~20 mg/L驴食草酚系列浓度筛选驴食草酚对HUVEC细胞毒作用。H_(2)O_(2)作用于HUVEC,评价驴食草酚对H_(2)O_(2)引起HUVEC损伤的保护作用。银杏叶提取物(GBE)为阳性对照药。倒置显微镜观察细胞形态,CCK-8法检测细胞存活率,丙酮酸还原法检测乳酸脱氢酶(LDH)释放量。结果随着H_(2)O_(2)浓度的增加,HUVEC的受损程度逐渐增加,细胞形态发生异常改变,细胞存活率逐渐降低,LDH释放量逐渐增加,综合确定H_(2)O_(2)损伤HUVEC浓度为750μmol/L。1.25~10 mg/L驴食草酚对HUVEC无细胞毒性,20 mg/L驴食草酚对HUVEC开始产生毒作用,最高无毒浓度为10 mg/L。驴食草酚可改善H_(2)O_(2)所致HUVEC细胞形态异常改变,提高HUVEC细胞存活率,减少LDH释放量。结论驴食草酚可有效减轻H_(2)O_(2)引起的HUVEC损伤。
Objective To investigate the protective effect of vestitol on human umbilical vein endothelial cell(s HUVEC)injury induced by hydrogen peroxide(H_(2)O_(2)).Methods HUVEC were cultured in vitro,and a series of concentrations of H_(2)O_(2)at 700~900μmol/L were set to screen the damaged concentration of H_(2)O_(2)to HUVEC.A series of concentrations of vestitol at 1.25~20 mg/L were set up to screen the cytotoxic effect of vestitol on HUVEC.HUVEC were exposed to H_(2)O_(2)to evaluate the protective effect of vestitol on HUVEC injury induced by H_(2)O_(2).Ginkgo biloba extract(GBE)was used as the positive control drug.The cell morphology was observed by the inverted microscope,the cell viability was detected by CCK-8 method and the lactate dehydrogenase(LDH)release was detected by the pyruvate reduction method.Results Along with the increase of H_(2)O_(2)concentration,the damage degree of HUVEC gradually increased,the cell morphology was injured,the cell viability gradually decreased,and the LDH release gradually increased.The damaged concentration of H_(2)O_(2)to HUVEC was determined to be 750μmol/L.1.25~10 mg/L vestitol had no cytotoxicity to HUVEC,20 mg/L vestitol began to produce toxic effects on HUVEC,and the highest non-toxic concentration was 10 mg/L.Vestitol can improve the abnormal morphological changes of HUVEC caused by H_(2)O_(2),increase the cell viability of HUVEC,and reduce the release of LDH.Conclusion Vestitol can effectively reduce HUVEC damage caused by H_(2)O_(2).
