摘要
结直肠癌是最常见的消化系统癌症之一,其晚期或转移性肿瘤患者的预后仍然很差。而胰岛素样生长因子结合蛋白4(IGFBP-4)是一种多功能的细胞增殖调控因子,具有IGFBP结构域和甲状腺球蛋白I型结构域,其在骨骼发育、细胞增殖、信号转导以及调控细胞生长方面发挥着重要的作用,并且参与生物体的衰老过程以及多种疾病的发生。在结直肠肿瘤中,IGFBP-4既可通过与胰岛素样生长因子(IGF)结合发挥抑制肿瘤细胞增殖、分化以及生长,又可通过Wnt/β-catenin、IL-6、Bcl-2/Bax等多种信号通路参与结直肠肿瘤发生与发展过程。因此,本研究就近年关于IGFBP-4结构功能以及其与结直肠肿瘤发生关系的研究进展进行分析和总结。
Colorectal cancer is one of the most common digestive system cancers,and the prognosis remains poor among patients with advanced or metastatic tumors.The insulin-like growth factor binding protein 4(IGFBP-4)is a multifunctional cell proliferation regulator with IGFBP domain and thyroglobulin type I domain,which plays an important role in bone development,cell proliferation,signal transduction and the regulation of cell growth.It has been shown to be involved in the aging process of organisms and the occurrence of many diseases.In colorectal tumors,IGFBP-4 can not only inhibit the proliferation,differentiation and growth of tumor cells by binding with insulin-like growth factor(IGF),but also can participate in the genesis and development of colorectal tumors through various signaling pathways,such as Wnt/β-catenin,IL-6,and Bcl-2/Bax.In light of this,the current study summarizes the recent research progress on the structure and function of IGFBP-4 and its relationship with the occurrence of colorectal tumors.
