摘要
目的筛选阿维链霉菌高产菌株。方法采用ARTP诱变和aveR基因过表达2种选育方式对阿维链霉菌进行复合筛选。结果经ARTP诱变筛选出RW-51诱变菌株效价为7268 U/mL,高于原始菌株8.9%。再结合aveR基因过表达菌株的构建,筛选出高产菌株RW-997初筛效价7649 U/mL,高于RW-51诱变菌株7.7%,高于原始菌株14.6%。结论筛选出1株高产菌株RW-997,用于大生产。
Objective The purpose of this study is to screen high-yield strains of Streptomyces avermitilis.Methods In this study,two breeding methods,ARTP mutation and aveR gene overexpression,were used to screen Streptomyces avermitilis.Results The RW-51 mutant strain used in this investigation had a titer of 7268 U/mL,which was 8.9%greater than the original strain after ARTP mutagenesis screening.When the aveR gene was constructed and overexpressed,a high-yield strain RW-997 was screened with a titer of 7649 U/mL,which was 14.6%greater than that of the original strain and 7.7%higher than that of the mutant strain RW-5.Conclusion One high-yielding strain RW-997 was selected for large-scale production.
