摘要
青榨槭为目前重要的秋季彩叶园林绿化优良树种之一。本研究基于青榨槭叶片转录组数据进行了SSR标记分析,结果发现其中3744条unigene序列中含有3763个SSR位点,发生频率为6.93%,每11.89 kb含一个位点。优势重复序列类型为二核苷酸重复和三核苷酸,分别占总SSR位点的51.21%与46.93%;AG/CT与GAA/TTC分别是二核苷酸和三核苷酸的优势重复基元。654对引物中252对(38.53%)引物可成功扩增出目的条带,75对(11.47%)引物表现出多态性。75对多态性引物在槭属16个种质间转移率范围为83.75%~100%,平均转移率为90.08%,表明这75对标记具有较高的通用性。利用转录组开发青榨槭SSR标记可为青榨槭以及槭属种质资源研究、分子标记辅助育种等提供重要的遗传资源。
Acer davidii is one of the most important tree species for autumn color leaf landscaping.In this study,SSR marker analysis was performed based on the transcriptome data of Acer davidii leaves.Among them,3 744unigene sequences contained 3 763 SSR sites,with a frequency of 6.93%,and each 11.89 kb contained one site.The dominant repeat sequence types are dinucleotide repeats and trinucleotide repeats,accounting for 51.21% and46.93% of the total SSR sites respectively;AG/CT and GAA/TTC are the dominant dinucleotide and trinucleotide repeats,respectively.Among 654 primer pairs,252 pairs(38.53%) of primers can successfully amplify the target band,and 75 pairs(11.47%) of primers showed polymorphism.The transfer rate of 75 pairs of polymorphic primers among 16 germplasms of Acer genus ranged from 83.75% to 100%,and the average transfer rate was90.08%,indicating that these 75 pairs of markers have high versatility.Using the transcriptome to develop SSR markers of A.davidii can provide important genetic resources for A.davidii and Acer germplasm resources research and molecular marker-assisted breeding.
