摘要
为探究INHBA基因在藏猪与大约克猪中的多态性与表达差异。在DNA水平上对藏猪、大约克猪INHBA基因5'侧翼区和CDS区进行了单核苷酸多态性(Single nucleotide polymorphism,SNP)位点筛选,同时在mRNA水平上用荧光定量PCR(qRT-PCR)方法检测INHBA基因在卵巢组织中的相对表达量。结果发现:INHBA基因在5'侧翼区存在一个突变位点A-1589T,并在藏猪与大约克猪2个品种间存在极显著差异(P<0.01);经转录因子预测,发现该突变位点与繁殖性能相关,并推测消失的转录因子结合位点可能是导致藏猪繁殖力低的主要原因之一。INHBA基因在卵巢中,藏猪表达量极显著高于大约克猪(P<0.01),推测INHBA基因可能与猪繁殖性能有关,并对提高猪的繁殖性能起到积极作用。本研究结果为进一步探究INHBA基因在猪繁殖性能的作用机制提供理论支撑。
This study was conducted to investigate the polymorphism and expression difference of INHBA gene in Tibetan and Yorkshire pigs.Single nucleotide polymorphism(SNP)sites were screened at the DNA level for the 5'flanking region and CDS region of INHBA gene in Tibetan and York pigs,and the relative expression of INHBA gene in ovarian tissues was measured at the mRNA level by fluorescent quantitative PCR(qRT-PCR).The results showed that there was a mutation site A-1589T in the 5'flanking region of INHBA gene,which exhibited significant difference(P<0.05)between Tibetan and Yorkshire pigs.Transcription factor prediction indicated an association between the mutation site and reproductive performance,suggesting that the absence of a transcription factor binding site may be one of the primary factors contributing to the reduced fertility observed in Tibetan pigs.Furthermore,the expression of the INHBA gene in ovarian tissues was significantly higher in Tibetan pigs compared to Yorkshire pigs (P<0.01), and it was hypothesized that the INHBA gene might be associated with and play a positive role in improving the reproductive performance of pigs. The results of this study provide theoretical support for further investigation of the mechanism of INHBA gene in pig reproductive performance. The findings of this study offer theoretical support for further investigations into the role of the INHBA gene in pig reproductive performance mechanisms.
