猪源芽孢杆菌的分离鉴定及其生物学特性研究

摘要 为了分离出潜在的猪源益生芽孢杆菌并探究其益生作用以期获得几株优良的芽孢杆菌菌株用于生产实践。试验采用仔猪粪便为原料,通过菌落形态观察、革兰氏染色、生理生化性质鉴定和16S rDNA序列分析、开展生物学特性分析、进行体外抑菌、药敏和淀粉酶、蛋白酶及纤维素酶活性测定等试验,对用营养琼脂固体培养基分离筛选出的两株菌株进行了研究。经过16S rDNA序列分析,确定培养得到的两株细菌为芽孢杆菌(Bacillus haynesii和Bacillus aerius),Bacillus aerius对大肠杆菌和金黄色葡萄球菌的生长繁殖没有明显的抑制作用;在试验所用到的十种药物中,这两株菌除了对红霉素不敏感外,对其余九种药物均有一定的敏感性;在酶活性试验中,两株菌的蛋白酶和淀粉酶活性比较强,纤维素酶的活性较弱。试验结果表明,这两株芽孢杆菌具有进一步研究并作为益生菌株开发利用的潜能。 In order to isolate potential porcine bacillus probiotics and explore its probiotic effects,and obtain several excellent bacillus strains serving for production practice,piglets stool test was used as raw material,through the colony morphology observation,gram staining,physiological and biochemical properties evaluation,16S rDNA sequence analysis,the biological characteristics analysis,in vitro antimicrobial susceptibility and amylase,protease,and determination of cellulose enzyme activity test,to study two strains form nutrient AGAR solid medium separation.Bacillus haynesii and Bacillus aerius were determined by 16S rDNA sequence analysis.Bacillus aerius had no obvious inhibitory effect on the growth and reproduction of Escherichia coli and Staphylococcus aureus.Among the ten drugs used in the experiment,these two strains were not sensitive to erythromycin,but were sensitive to the other nine drugs.In enzyme activity test,the protease and amylase activities of the two strains were stronger,while the cellulase activity was weaker.The results showed that these two strains had the potential to be further studied and exploited as probiotic strains.
