m6A甲基化甲基转移酶RBM15在宫颈癌中的表达及临床意义

摘要 目的探究m6A甲基化甲基转移酶RNA结合蛋白15(RNA-binding Motif Protein 15,RBM15)与宫颈癌的相关性及临床意义。方法选取2011年1月1日至2018年1月1日期间于皖南医学院第一附属医院进行诊治的宫颈癌病例132例,另选20例正常宫颈组织病例作为对照,共152例,通过免疫组化法检测RBM15在宫颈癌中和正常宫颈中的表达水平,分析其相关性,并研究其表达量与临床分期及临床病理特征之间的关系。结果RBM15在宫颈癌中的阳性表达率56%显著高于正常宫颈0%(P=0.001),且宫颈癌的期别越高,RBM15表达越强(P=0.002)。复发宫颈癌中RBM15的表达也呈现为随期别逐渐上升的趋势(P=0.156)。RBM15的阳性表达与宫颈癌的肿瘤最大径、淋巴结转移、神经侵犯、脉管癌栓有关(P<0.05),而与肿瘤分级、病理类型、发病年龄、鳞状上皮细胞癌抗原(Squamous cell carcinoma antigen,SCCA)无关(P>0.05)。结论RBM15在宫颈癌中呈高表达,且与宫颈癌分期及临床病理特征具有一定的相关性,RBM15表达升高也会造成宫颈癌患者预后不良,这一发现日后有望成为宫颈癌诊治的新靶点。 Objective To investigate the correlation between m6A methylated methyltransferase RNA-binding Motif Protein 15(RBM15)and cervical cancer and its clinical significance.Methods A total of 132 cases of cervical cancer diagnosed and treated in the First Affiliated Hospital of Wannan Medical College from Jan 1st 2011 to Jan 1st 2018 were selected,and 20 cases of normal cervical tissue were selected as control.A total of 152 cases were detected by immunohistochemical method to detect the expression of RBM15 in cervical cancer and normal cervix,and the correlation was analyzed.The relationship between RBM15 expression and clinical stage or clinicopathological features was studied.Results The expression of RBM15(56%)in cervical cancer was significantly higher than that in normal cervix(0%)(P<0.05),and the higher the stage of cervical cancer was associated with the higher expression of RBM15(P<0.05).The expression of RBM15 in recurrent cervical cancer also showed a gradually increasing trend with the period(P=0.156>0.05).Positive expression of RBM15 was associated with maximum tumor diameter,lymph node metastasis,nerve invasion and vascular cancer thrombus(P<0.05),but not with tumor grade,pathological type,age of onset and squamous cell carcinoma antigen(SCCA)(P>0.05).Conclusion RBM15 is highly expressed in cervical cancer,and has a certain correlation with the stage and clinicopathological characteristics of cervical cancer.The increased expression of RBM15 will also cause poor prognosis of cervical cancer patients,which is expected to be a new target for the diagnosis and treatment of cervical cancer in the future.
