电针对反复丙泊酚暴露大鼠炎症反应的影响

摘要 目的:观察电针对反复丙泊酚暴露SD大鼠炎症反应的影响。方法:SPF级SD大鼠40只,日龄10 d,体质量26~28 g,采用随机数字表法将其分为5组(每组8只):生理盐水对照(C)组,腹腔注射生理盐水100μL;脂肪乳剂(F)组,腹腔注射脂肪乳剂100μL;电针(E)组,选取大鼠“百会穴”“大椎穴”进行电针刺激,疏密波2 Hz/10 Hz,30 min/次,1次/d,连续3 d;丙泊酚(P)组,腹腔注射异丙酚100 mg/kg;电针+丙泊酚(EP)组,先给予丙泊酚(同P组),后电针干预(同E组)。造模结束后,断头取大鼠海马组织。Western blot法检测海马组织中白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)表达量。免疫荧光检测海马小胶质细胞高迁移率族蛋白B1(HMGB1)、离子钙结合衔接分子1(Iba-1)表达水平。结果:与C组比较,P组海马组织中TNF-α、IL-6表达量升高,差异具有统计学意义(P<0.05);海马小胶质细胞Iba-1、HMGB1表达水平明显升高,差异具有统计学意义(P<0.05)。与P组比较,EP组海马组织中TNF-α、IL-6表达量下降,差异具有统计学意义(P<0.05);海马小胶质细胞Iba-1、HMGB1表达水平明显下降,差异具有统计学意义(P<0.05)。结论:电针干预能减轻反复丙泊酚暴露的大鼠神经炎症反应,其部分机制可能是抑制海马组织中小胶质细胞的活化和坏死性凋亡,下调炎症因子TNF-α、IL-6水平。 Objective:To investigate impact of electro-acupuncture(EA) on propofol-induced inflammatory response.Methods:A total of 40 SD rats(10 d,26 g-28 g) were randomly divided into the control group,the fat emulsion group,the EA group,the propofol group and the combination group,with 8 rats in each group.The control group was given intraperitoneal injection of normal saline(100 μL);the fat emulsion group was given intraperitoneal injection of fat emulsion(100 μL);the EA group was intervened with electro-needling Baihui(DU20) and Dazhui(DU14),dilatational wave of 2 Hz/10 Hz,30 minutes daily for 3 days;the propofol group was treated with propofol injection(100 mg/kg);the combination group was treated with EA plus propofol injection.Expressions of interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) were detected by Western blot.The levels of high mobility group protein B1(HMGB1) and ionized calcium binding adapter molecule 1(Iba-1) in microglia of hippocampus were detected by immunofluorescence.Results:The expressions of TNF-α and IL-6 in the hippocampus were higher(P<0.05),and the levels of Iba-1 and HMGB1 in the microglia of hippocampus were significantly increased in the propofol group compared with those in the control group(P<0.05).Compared with those in the propofol group,the expressions of TNF-α and IL-6 were decreased(P<0.05),and the levels of Iba-1 and HMGB1 were significantly reduced in the combination group(P<0.05).Conclusion:EA intervention can inhibit the activation and the necroptosis of microglia in the hippocampus,and down-regulate the levels of TNF-α and IL-6,which leads to a reduction in the repeated propofol-induced neuroinflammatory response in rats.
