向日葵油、山茶油和亚麻子油对脂多糖诱导HaCaT细胞的作用

Effects of Sunflower Seed Oil,Camellia Oil and Linseed Oil on Lipopolysaccharide-Induced HaCaT Cells

摘要 为研究向日葵油、山茶油和亚麻子油对损伤的角质形成细胞的修复作用机制,构建脂多糖(LPS)诱导的人永生化角质形成细胞(HaCaT细胞)作为损伤研究模型,通过免疫印迹法和实时定量荧光PCR(qPCR)分别检测植物油对细胞损伤模型中丝裂原活化的蛋白激酶(MAPKs)信号通路的磷酸化激活水平和屏障相关蛋白mRNA表达情况。结果发现在1%(质量分数)时山茶油和亚麻子油能够显著抑制MAPKs信号通路主要蛋白的磷酸化水平和人组织激肽释放酶7(KLK7)mRNA的表达,显著促进丝聚蛋白(FLG)和半胱氨酸天冬氨酸特异性蛋白14(Caspase14)mRNA表达,提示山茶油和亚麻子油可抑制MAPKs信号通路介导的生物学损伤,促进LPS诱导的HaCaT细胞修复。 In order to study the repair mechanisms of sunflower seed oil,camellia oil and linseed oil on damaged keratinocytes,lipopolysaccharide(LPS)-induced human immortalized keratinocytes(HaCaT cells)were used as the study model.Immunoblotting and real-time fluorescence quantitative polymerase chain reaction(qPCR)were used to detect the phosphorylation levels of mitogen activated protein kinases(MAPKs)signaling pathway and the mRNA expression of barrier associated protein.The results showed that camellia oil and linseed oil could significantly inhibit the phosphorylation of major proteins in MAPKs signaling pathway and kalkinase 7(KLK7)mRNA expression,and also significantly increased the expression of filagmin(FLG)and Caspase14 mRNA at a mass fraction of 1%.These results suggest that camellia oil and linseed oil,in particular,can inhibit the biological damage mediated by MAPKs signaling pathway and promote LPS-induced HaCaT cells repair.
