摘要
COL6A3是COL6家族的一员,在多个生物过程中发挥关键作用,已被报道与多种恶性肿瘤的进展相关.然而,它在胃癌中的作用仍不清楚.在这项研究中,通过分析TCGA数据库,发现COL6A3 mRNA在胃癌组织中上调,并与胃癌患者的预后呈现负相关.通过分析我院收集的胃癌组织,发现COL6A3在人类胃癌组织中高表达.数据进一步证实了COL6A3的表达与胃癌患者的临床病理特征包括肿瘤大小(P=0.006)之间的关联性.在人类胃癌细胞系MGC-803和AGS中,通过CCK-8和细胞集落形成实验发现敲低COL6A3能抑制细胞增殖,动物实验结果表明,敲低COL6A3可以明显减小小鼠的肿瘤体积.综上所述,COL6A3可能成为治疗胃癌的新靶点和预后标志物.
COL6A3,a member of the COL6 family,plays a key role in multiple biological processes and has been reported to be associated with the progression of multiple malignancies.However,its role in gastric cancer remains unclear.By analyzing the TCGA database,it was found that COL6A3 m RNA is upregulated in cancer tissue and negatively correlated with the prognosis of cancer patients.Through the analysis of gastric cancer tissues collected in our hospital,we found that COL6A3 was highly expressed in human gastric cancer tissues.The correlation between COL6A3 expression and clinical pathological features,including tumor size,in cancer patients was further confirmed by our data(P=0.006).In human gastric cancer cell line MGC-803and AGS,we found that knocking out COL6A3 inhibited cell proliferation through CCK-8 and cell colony formation tests,and animal experiments showed that knocking down COL6A3 could significantly reduce tumor volume in mice.COL6A3 may be a new target and prognostic marker for the treatment of gastric cancer.
