长非编码RNA NRAV在泛癌中的表达和功能研究

摘要 LncRNA NRAV是近年来新发现的lncRNA分子,仅少数研究显示其表达水平与肺癌、胰腺导管腺癌的发展紧密相关,但NRAV在其它癌症中的表达情况和功能尚不清晰。通过生物信息学的方法分析NRAV在不同癌症中的差异表达情况及其与癌症患者生存期的关系,并以肝细胞癌(LIHC)作为重点研究对象,基于转录组数据筛选与NRAV共表达的mRNA并进行GO和KEGG功能富集分析,探究NRAV在LIHC中的调控机制与功能。结果显示,NRAV在7种癌症与癌旁组织中差异表达显著,其表达量与5种癌症患者的总生存期以及4种癌症患者的无病生存期显著相关。另外,筛选出的479个共表达靶基因主要富集在DNA修复、蛋白质结合、细胞周期等通路上。该文为进一步了解NRAV与泛癌尤其是肝细胞癌的发生发展关系提供初步依据。 The lncRNANRAV,which has been recently discovered,exhibits a close correlation with the progression of lung cancer and pancreatic ductal adenocarcinoma.However,the expression and function of NRAV in other cancers are still unclear.In this study,we analyze the differential expression of NRAV in different cancers and their influence on the survival of different patients based on the bioinformatics methods.Subsequently,we select the targets genes of NRAV in liver hepatocellular carcinoma(LIHC)for further analysis.To evaluate the regulatory mechanism and function of NRAV in LIHC,the GO(Gene Ontology)and KEGG(Kyoto encyclopedia of genes and genomes)analyses of the NRAV co-expression genes are performed.The findings reveal significant differential expression of NRAV across seven different types of cancer as well as their corresponding normal tissues.Moreover,it demonstrates a significant correlation between NRAV expression levels and overall survival rates among five types of cancer patients,as well as disease-free survival rates among four types of cancer patients.Notably,the 479 co-expressed mRNA are mainly enriched in DNA repair,protein binding,cell cycle and other pathways.This study provides an initial foundation for comprehending the intricate relationship between NRAV and the initiation and progression of pan-cancer.
