摘要
产自红色邻米草菌株S2-4-1H^(T)的六氢环庚基灵菌红素(six hydrogen cycloheptane prodigiosin,ShcPG)具有极高的抗肿瘤新药开发潜力。为提高摇瓶发酵产ShcPG的产量,试验的菌株为红色邻米草菌S2-4-1H^(T),基础培养基为MB,以庚基灵菌红素(heptylprodigiosin,hPG)和ShcPG质量浓度为响应值,在单因素试验基础上采用Box-Behnken中心组合试验设计进行响应面试验,优化S2-4-1H^(T)菌株发酵条件,并构建二次回归方程。结果表明,发酵优化条件为温度28℃、pH 7.73和总盐度2.85%。在最佳发酵条件工艺下发酵48 h,hPG和ShcPG的总产量为5.71±0.12 mg/L,是用MB培养基培养产量的22.84倍,且预测值和测定值的绝对差值<5%,表明回归模型均可靠。该结果对红色邻米草菌S2-4-1H^(T)发酵开发生产Shc PG具有实际应用价值。
The six hydrogen cycloheptane prodigiosin(ShcPG) from Spartinivicinus ruber strain S2-4-1H^(T) has high potential for development of antitumor drugs.In order to improve the yield of ShcPG from S.ruber strain S2-4-1H^(T) by shaking flask fermentation,a response surface experiment adopting the Box-Behnken central composite design was carried out following single factor experiments.In this study,S.ruber strain S2-4-1H^(T) was took as the test strain,MB medium as the base medium and the concentration of the heptylprodigiosin(hPG) and ShcPG as the response value.The medium components and conditions of S.ruber strain S2-4-1H^(T) fermentation were optimized,and the quadratic regression equation was established.The results showed that the optimal fermentation conditions were as follows:temperature,pH and total salinity were 28 ℃,7.73 and 2.85%,respectively.Under the optimal fermentation conditions,the fermentation time was 48 h,and the yield of Shc PG was 5.71±0.12 mg/L,which was 22.84 times higher than that cultured in MB medium.Under these two conditions,the absolute difference of ShcPG yield between the predicted value and the measured value was less than 5%,indicating that the regression model was reliable.These results have practical application value for the development and production of ShcPG by S.ruber strain S2-4-1H^(T) fermentation.
