不同生长年限九节茶不同部位中八种成分含量比较

摘要 目的建立高效液相色谱法同时测定不同生长年限九节茶不同部位中的8种成分含量变化。方法色谱柱为Phenomenex Luna(C18)(250 mm×4.6 mm,5μm),以0.2%磷酸(A)-乙腈(B)为流动相,梯度洗脱(0~12 min,2%~11%B;12~25 min,11%~18%B;25~40 min,18%~40%B),流速1.0 ml/min,检测波长为210 nm和325 nm,柱温30℃,进样量10μl。结果富马酸、新绿原酸、绿原酸、隐绿原酸、咖啡酸、异嗪皮啶、落新妇苷、迷迭香酸8种成分分别在7.61~304.40μg/ml(R^(2)=0.9997)、3.52~140.60μg/ml(R^(2)=1.0000)、4.99~199.60μg/ml(R^(2)=1.0000)、3.89~155.40μg/ml(R^(2)=1.0000)、0.56~22.06μg/ml(R^(2)=0.9999)、0.57~22.62μg/ml(R^(2)=1.0000)、26.75~1070.00μg/ml(R^(2)=1.0000)、12.49~499.40μg/ml(R^(2)=0.9999)范围内线性关系良好。不同添加水平加标回收率为95.05%~99.26%,其相对标准偏差在1.15%~1.93%之间。结论九节茶在茎中富马酸、绿原酸、咖啡酸、落新妇苷、迷迭香酸的含量1年生>2年生>3年生;新绿原酸、隐绿原酸的含量1年生<2年生<3年生;异嗪皮啶的含量1年生>3年生>2年生。在叶中富马酸、新绿原酸、隐绿原酸、咖啡酸、异嗪皮啶、落新妇苷、迷迭香酸的含量1年生>2年生>3年生;绿原酸的含量1年生>3年生>2年生。 Objective To establish a HPLC method for simultaneous determination of eight components in different growth years and different parts of Sarcandra glabru.Methods The Phenomenex Luna C18(250 mm×4.6 mm,5μm)column was used,the mobile phase was 0.2%phosphoate acid(A)and acetonitrile(B),gradient elution(0~12 min,2%~11%B;12~25 min,11%~18%B;25~40 min,18%~40%B)was performed;the flow rate was 1.0 ml/min;the detection wavelength was 210 nm and 325 nm,the column temperature was 30℃,the injection volume was 10μl.Results Within the following ranges,the linear relationship of eight components were good respectively:7.61~304.40μg/ml for fumaric acid(R^(2)=0.9997),3.52~140.60μg/ml for neochlorogenic acid(R^(2)=1.0000),4.99~199.60μg/ml for chlorogenic acid(R^(2)=1.0000),3.89~155.40μg/ml for cryptochlorogenic acid(R^(2)=1.0000),0.56~22.06μg/ml for caffeic acid(R^(2)=0.9999),0.57~22.62μg/ml for isofraxidin(R^(2)=1.0000),26.75~1070.00μg/ml for astilbin(R^(2)=1.0000),12.49~499.40μg/ml(R^(2)=0.9999)for rosmarinic acid(R^(2)=1.0000),the recovery rate of different addition levels was 95.05%~99.26%,and its relative standard deviation was between 1.15%and 1.93%.Conclusion In the stem,the content of fumaric acid,chlorogenic acid,caffeic acid,astilbin,rosmarinic acid were 1 year>2 years>3 years;the content of neochlorogenic acid,cryptochlorogenic acid were 1 year<2 years<3 years;the content of isofraxidin was 1 year>3 years>2 years.In the leaves,the content of fumaric acid,neochlorogenic acid,cryptochlorogenic acid,caffeic acid,isofraxidin,astilbin,rosmarinic acid were 1 year>2 year>3 years;the content of chlorogenic acid was 1 year>3 years>2 years.
