党参多糖的抗氧化活性研究及其对蛋鸡血清免疫、抗氧化功能的影响

摘要 试验旨在探究党参多糖(CPP)抗氧化活性及其对蛋鸡血清免疫、抗氧化功能的影响。研究通过体外试验检验CPP对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、羟自由基和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS^(+))自由基的清除能力,体内试验检验CPP对蛋鸡血清新城疫抗体效价、抗氧化酶活性的影响。选取1日龄健康罗曼蛋鸡40羽,随机分为4组,每组10个重复,每个重复1羽蛋鸡,分别为CPP高浓度组(CPP_(H)组)、CPP低浓度组(CPPL组)、免疫对照组(VC组)、空白对照组(BC组)。试验期35d。结果显示,CPP对DPPH自由基、羟自由基和ABTS^(+)自由基均具有不同程度的清除能力,且随着CPP浓度的增加,其抗氧化活性显著增强。首免后7、21d,CPP_(H)组血清过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性及新城疫抗体效价显著高于VC组和BC组(P<0.05)。研究表明,CPP的抗氧化活性较强,能够提高蛋鸡血清抗氧化酶活性及新城疫抗体效价。 The purpose of this study was to explore the antioxidant activity of Codonopsis polysaccharide(CPP)and its effect on serum immunity and antioxidant function of laying hens.In vitro tests were used to test the scavenging ability of CPP on DPPH free radicals,hydroxyl free radicals and ABTS^(+)free radicals,and in vivo tests were used to examine the effects of CPP on laying hens serum Newcastle disease antibody titer and antioxidant enzyme activities.Forty one-day-old healthy Roman laying hens were randomly divided into four groups with 10 replicates per group and one laying hen per replicate.Laying hens were divided into CPP high concentration group(CPP_(H) group),CPP low concentration group(CPPL group),immune control group(VC group)and blank control group(BC group),respectively.The experiment lasted for 35 days.The results showed that CPP had different degrees of scavenging ability against DPPH free radicals,hydroxyl radicals and ABTS^(+)free radicals,and its antioxidant activity was significantly enhanced with the increase of CPP concentration.At 7 and 21 days after the first immunization,the serum CAT,GSH-Px activities,and Newcastle disease antibody titers in the CPP_(H) group were significantly higher than those in the VC group and BC group(P<0.05).The results showed that CPP had strong antioxidant activity,which could improve the serum antioxidant enzyme activities and Newcastle disease antibody titer of laying hens.
