水稻耐盐性基因SKC1的KASP标记的开发与利用

摘要 籼稻品种‘Nona Bokra’中携带的SKC1^(NB)等位基因水稻耐盐性的主效QTL(Quantitative trait locus)位点。为了提高对水稻SKC1基因型鉴定效率,根据SKC1^(NB)基因中的功能性SNP位点开发了一个KASP分子标记SKC1^(NB)-KASP,利用该标记对‘Nona Bokra’与粳稻品种‘繁14’、‘花B’和‘武1B’的回交BC3F2群体进行SKC1等位基因的分型,发现在BC3F2群体受测的65个单株的DNA样品中,有16份样品的基因型与供体亲本‘Nona Bokra’相同,31份样品显示杂合基因型,18份样品与轮回亲本基因型相同。对这65份DNA样品的,SKC1基因进行测序以检验KASP标记基因分型结果的准确性,发现测序结果与KASP标记分型的结果完全一致。以上结果说明SKC1^(NB)-KASP标记可以高效、准确地鉴定,SKC1位点的基因型,大大提高了耐盐性水稻分子标记辅助选择育种的效率。 SKC1^(NB)allele in indica rice variety'Nona Bokra',is a major QTL for salt-tolerant in rice.In order to improve the genotype screening efficiency for SKC1 locus,a KASP marker,SKC1^(NB)-KASP was developed based on the functional SNP site of the SKC1^(NB)allele.Genotyping of 3 backcross population,which constructed with‘Nona Bokra’as donor parent and Fan 14,HuaB and WulB as recurrent parent,respectively,indicated that among the 65DNA samples of BC3F2 population,16 samples had the same genotype as the donor parent ‘Nona Bokra’,31 samples showed heterozygous genotype,and 18 samples had the same genotype as the recurrent parents.Subsequently,the SKC1 gene of these 65 DNA samples was sequenced to verify the accuracy of the KASP marker genotyping results,and it was found that the sequencing results were completely consistent with the KASP marker genotyping results.These results indicate that the SKC1^(NB)-KASP marker can efficiently and accurately screen genotype for SKC1 loci,and robust enough for molecular marker-assisted selection breeding for salt-tolerant rice.
