摘要
目的探究三结构域蛋白59(TRIM59)、细胞周期蛋白E1(CCNE1)在宫颈癌组织中的表达及二者与宫颈癌患者预后的关系。方法选择2015年1月至2017年1月在大连市妇女儿童医疗中心接受治疗的宫颈癌患者120例,术中取其癌组织和癌旁组织分别作为观察组和对照组。采用免疫组织化学法测定TRIM59、CCNE1表达。采用Kaplan-Meier法分析TRIM59、CCNE1表达与患者预后的关系;采用多因素COX回归分析影响宫颈癌患者预后的危险因素。结果观察组TRIM59、CCNE1阳性表达率比对照组升高(P<0.05)。宫颈癌患者TRIM59、CCNE1表达与患者年龄、肿瘤直径、病理类型无关(P>0.05),与患者TNM分期、分化程度和淋巴结转移有关(P<0.05)。Kaplan-Meier生存曲线分析显示,宫颈癌患者TRIM59阳性表达组生存率低于阴性表达组(χ^(2)=4.901,P=0.027);CCNE1阳性表达组生存率低于阴性表达组(χ^(2)=5.583,P=0.018)。TRIM59阳性表达、CCNE1阳性表达、淋巴结转移、低度分化、TNM分期(Ⅱa)是影响宫颈癌患者发生不良预后的危险因素(P<0.05)。结论宫颈癌患者癌组织中TRIM59、CCNE1表达异常升高,二者均为宫颈癌患者发生不良预后的危险因素。
Objective To explore the expression of tripartite motif-59(TRIM59)and cyclin E1(CCNE1)in cervical cancer tissues and their relationship with the prognosis of cervical cancer patients.Methods A total of 120 patients with cervical cancer who were admitted to Dalian Women and Children Medical Center(Group)from January 2015 to January 2017 were selected.The cancer tissues and adjacent tissues were taken intraoperatively as the observation group and the control group respectively.The expression of TRIM59 and CCNE1 was determined by immunohistochemistry.Kaplan-Meier method was used to analyze the relationship between the expression of TRIM59 and CCNEl and the prognosis of patients;and multivariate COX regression analysis was used to analyze the risk factors affecting the prognosis of cervical cancer patients.Results The positive expression rates of TRIM59 and CCNE1 in the observation group were higher than those in the control group(P<0.05).The expression of TRIM59 and CCNE1 in cervical cancer patients was not related to patient age,tumor diameter and pathological type(P>0.05),but was related to TNM stage,differentiation degree and lymph node metastasis(P<0.05).Kaplan-Meier survival curve analysis showed that the survival rate of cervical cancer patients with TRIM59 positive expression group was lower than that of negative expression group(χ^(2)=4.901,P=0.027);the survival rate of CCNEI positive expression group was lower than that of negative expression group(χ^(2)=5.583,P=0.018).TRIM59 positive expression,CCNEl positive expression,lymph node metastasis,poorly differentiated,TNM stage(a)were risk factors for poor prognosis in cervical cancer patients(P<0.05).Conclusion The expression of TRIM59 and CCNE1 in cervical cancer tissues is abnormally increased,both are risk factors for poor prognosis in patients with cervical cancer.
