摘要
袖状胃切除术,又称缩胃手术,主要用于减重和2型糖尿病的治疗。为了探究袖状胃切除术对大鼠糖代谢及生长调控相关激素Ghrelin、IGF-Ⅰ的影响。本研究将试验组SD大鼠行袖状胃切除术,对照组行假手术。术后第7天,检测血浆中Ghrelin、IGF-Ⅰ、葡萄糖含量及脏器组织样本中的IGF-Ⅰ、IGFBP-3、GHS-R及GH-R的m RNA表达量。试验组术后血浆中Ghrelin、IGF-Ⅰ水平极显著低于对照组(P<0.01),肝脏、肾脏及肺脏的IGF-Ⅰ和GH-R的m RNA表达量均极显著低于对照组(P<0.01),肝脏的IGFBP-3和肾脏的GHS-R的m RNA也极显著低于对照组(P<0.01)。此外,试验组术后血糖水平极显著下降(P<0.01),胰岛素水平也极显著低于对照组(P<0.01)。本研究结果表明袖状胃切除后大鼠Ghrelin、IGF-Ⅰ等激素水平显著下降,同时伴随着血糖显著下降,提示袖状胃切除导致的激素水平变化可能介导调控机体糖代谢。
Sleeve gastrectomy,also known as gastric reduction surgery,is mainly used for weight loss and the treatment of type 2 diabetes. This study aimed to investigate the effect of sleeve gastrectomy on glucose-metabolism and growth-regulating hormones Ghrelin and IGF-I in rats. In this study,the SD rats in the experimental group underwent sleeve gastrectomy,and the control group underwent sham surgery. On the 7th postoperative day,the plasma contents of ghrelin,IGF-Ⅰand glucose and the m RNA expression level of IGF-Ⅰ,IGFBP-3,GHS-R and GH-R were measured in organ tissue samples.The levels of ghrelin and IGF-Ⅰ in plasma of experimental group after surgery were significantly lower than those of control group(P<0.01),and the expression levels of IGF-Ⅰand GH-R m RNA in the liver,kidney and lung were significantly lower than those of control group(P<0.01).The m RNA levels of IGFBP-3 in liver and GHS-R in kidney were also significantly lower than those in control group(P<0.01).In addition,the postoperative blood glucose level of the experimental group decreased significantly(P<0.01),and the insulin level was significantly lower than that of control group(P<0.01).The results of this study showed Ghrelin,IGF-I and other hormones decreased significantly in rats after sleeve gastrectomy,accompanied by a significant decrease in blood glucose,suggesting that the changes in hormone levels caused by sleeve gastrectomy may affect mediate the regulation of glucose metabolism.
