摘要
为研究生殖衰老对小鼠卵泡颗粒细胞(Granulosa cells,GCs)发育的影响,本试验对课题组前期的转录组与蛋白质组数据进行了分析,这些数据来自于6周龄(年轻组)、10月龄(衰老组)的健康雌性昆明(KM)小鼠。结果显示,转录组数据与蛋白组数据得到的差异基因与差异蛋白的符合程度并不高,共筛选出95个基因和蛋白同时在转录水平和蛋白水平发生上调,有22个基因在转录水平和蛋白水平发生下调,采用qPCR与Westernblot方法对其中的相关基因的表达水平进行了验证。结果表明,与年轻组相比,衰老组卵泡颗粒细胞的凋亡相关基因Ndrg1和Sting1表达显著上升(P<0.05),抗氧化相关基因Gpx7、生长因子相关基因Gdf9、Bmpr1b与激素合成、分泌相关基因Esr2、Lhcgr表达显著下降(P<0.05)。综上说明机体发生生殖衰老后卵泡颗粒细胞的凋亡水平上升,抗氧化功能下降,激素作用途径受损。
In order to investigate the effects of aging on the development of Granulosa cells(GCs)in mouse follicles,we analyzed the transcriptomic and proteomic data from healthy female Kunming(KM)mice aged 6 weeks(young group)and 10 months(old group).The results showed that the differentially expressed genes were not highly consistent with the differentially expressed proteins.A total of 95 genes were up-regulated at both mRNA and protein levels,while 22 genes were down-regulated.The expression levels of related genes were verified by qPCR and Western blot.The results showed that the expressions of apoptosis-related genes Ndrg1 and Sting1 in the aging group were significantly increased(P<0.05)compared with the young group.Significant decreases(P<0.05)were found in antioxidant related genes Gpx7,growth factor related genes Gdf9,Bmpr1b and hormone synthesis and secretion related genes Esr2 and Lhcgr.In conclusion,the apoptosis level of follicular granulosa cells increased,the antioxidant function decreased,and the hormone action pathway was damaged after reproductive senescence.
