基于Illumina MiSeq高通量测序技术解析六堡茶产品中真菌群落特征

Analysis of fungal community characteristics of Liupao tea based on Illumina MiSeq high throughput sequencing technology

摘要 试验采用Illumina MiSeq高通量测序技术对七份不同发酵工艺六堡茶样品的真菌菌落结构及多样性进行解析,扩增真菌的ITS序列。试验结果表明,样品中真菌多样性测序共获得532,864条序列,125,975,232碱基,97操作分类单(operational taxonomic units,OUT),可以归为3个门、11个纲、23个目、34个科、44个属、60个种,其中在门水平上子囊菌属门(Ascomycota)为优势菌门,相对丰度90.61%~99.24%。在属水平上曲霉属(Aspergillus)为优势菌,相对丰度5.74%~99.89%。通过主坐标分析显示茶果(CG)、老茶婆(LCP)与其他五份样本之间真菌结构差异较大,两个主坐标叠加解释度达到73.73%;经过属水平热图分析显示老茶婆(LCP)和社前茶(SQC)两个样本具有较相似的物种组成。本文为六堡茶后发酵工艺开发与研究提供理论依据。 Seven samples of Liupao tea under different fermentation processes were analyzed by Illumina MiSeq high-throughput sequencing technology,in order to analyze the fungal community diversity and amplify the ITS sequence of fungi.The results showed that 532,864 sequences,125,975,232 bases,and 97 operational taxonomic units(OUT)were obtained,which could be classified into 3 phyla,11 classes,23 orders,34 families,44 genera,and 60 species.At the phylum level,Ascomycota was the dominant phylum with a relative abundance of 90.61%~99.24%.At the genus level,Aspergillus was the dominant bacterium,with relative abundance of 5.74%~99.89%.The results of principal coordinate analysis showed that the fungal structure of tea fruit(CG)and old tea woman(LCP)was significantly different from the other five samples,and the superposition explanation of the two principal coordinates reached 73.73%.Genus level heat map analysis showed that LCP and Sheqian tea(SQC)samples had similar species composition.This paper provides theoretical basis for the development and research of Liupao tea post-fermentation process.
