酸枣仁汤改善间氯苯哌嗪致小鼠焦虑样行为的作用

Roles of Suanzaoren Decoction in Improving Anxiety-like Behavior Induced by M-chloropiperazine in Mice

目的比较不同配方酸枣仁汤对单次腹腔注射间氯苯哌嗪致小鼠焦虑样行为的改善作用。方法不同配方酸枣仁汤给药7 d,采用高架十字迷宫实验检测其对间氯苯哌嗪诱导的小鼠焦虑样行为的改善作用;采用ELISA方法检测小鼠血清5-羟色胺、皮质酮、促肾上腺皮质激素以及脑中5-羟色胺的含量。结果在高架十字迷宫实验中,单次腹腔注射间氯苯哌嗪4 mg·kg^(-1)能够引起小鼠焦虑样行为,酸枣仁汤原方21.7 g·kg^(-1)可显著延长小鼠开臂停留时间,增加开臂停留时间的百分比,明显上调其血清中5-羟色胺含量,并使下丘脑、杏仁核、海马、皮层中5-羟色胺含量有降低的趋势,但未影响血清促肾上腺皮质激素和皮质酮含量。结论酸枣仁汤原方可改善单次腹腔注射间氯苯哌嗪诱导的小鼠焦虑样行为,其机制可能在于调节其5-羟色胺能神经系统。

Objective To compare the effect of different formulations of Suanzaoren decoction on anxiety-like behavior of mice induced by single intraperitoneal injection of m-chlorpiperazine.Methods After seven days of administration of different formulations of Suanzaoren decoction,the extent to which Suanzaoren decoction improved the anxiety-like behavior induced by m-chlorpiperazine was detected via the elevated-plus maze test.The contents of serum 5-hydroxytryptamine、corticosterone、adrenocorticotropic hormone and 5-hydroxytryptamine in the brain were detected by ELISA.Results In the elevated-plus maze test,single intraperitoneal injection of m-chlorpiperazine 4 mg·kg^(-1) induced anxiety-like behavior in mice.Suanzaoren decoction 21.7 g·kg^(-1) could prolong the time spent in open arms,increase the percentage of the time spent in open arms,and upregulate the content of 5-hydroxytryptamine in serum but downregulate the content of 5-hydroxytryptamine in the hypothalamus,amygdala,hippocampus and cortex.Serum adrenocorticotropic hormone and corticosterone were not affected by Suanzaoren decoction.Conclusion Suanzaoren decoction can improve anxiety-like behavior induced by single intraperitoneal injection of m-chlorpiperazine,and the mechanism may be related to regulating the serotonergic nervous system.
