摘要
羊肺炎支原体(Mo)可引起山羊、绵羊发生支原体肺炎。本研究旨在分析Mo感染对贵州不同品种羊Toll样受体(TLRs)MyD88通路因子基因转录水平的影响,进而分析不同品种羊对Mo感染的差异性。本研究以Mo人工感染贵州5个主要品种羊,应用TaqMan RT-qPCR方法对不同品种试验羊肺脏和血液样本TLRs MyD88通路因子基因转录水平进行定量检测与分析。结果显示,不同品种羊感染Mo后,MyD88、TRAF6、NF-κB、FOS、JUN基因mRNA相对表达量均显著上调(P<0.01)。同时发现,感染死亡的贵州黑山羊、贵州白山羊血液和肺脏样本中MyD88、FOS基因转录水平上调倍数均显著高于感染未死亡的波尔山羊、湖羊(P<0.05);感染未死亡的波尔山羊血液中NF-κB基因转录水平上调倍数均极显著高于其余四种羊(P<0.01),这种差异表达出现的原因可能是Mo对不同品种羊的致病机理不同。以上研究结果表明,贵州不同品种羊对Mo的易感程度与TLRs MyD88通路因子转录水平有关,为研究Mo对于不同品种羊的致病机制提供基础研究资料。
Mycoplasmaovipneumoniae(Mo)can cause pneumonia in goats and sheep.The transcript levels of MyD88 pathway factor genes of Toll-like receptors(TLRs)was analyzed in the present study to determine the effect on susceptibility of Mo infection in different breeds of sheep and goats in Guizhou.Five major breeds of sheep and goats in Guizhou were experimentally infected with Mo and the transcript levels of TLRs MyD88 pathway factors in lung and blood samples were quantified and analyzed by using TaqMan RT-qPCR.The results showed that the relative mRNA expression of MyD88,TRAF6,NF-κB,FOS and JUN genes were all significantly up-regulated after Mo infection in different breeds of sheep and goats(P<0.01).Meanwhile,the up-regulated transcript levels of MyD88 and FOS genes were significantly higher in the blood and lung samples of Guizhou black goats and Guizhou white goats that were infected and died than those of Boer goats and Lake sheep that were not infected and died(P<0.05).In addition,the up-regulation of NF-κB gene transcript levels in the blood of infected non-dead Boer goats were all very significantly higher than in the remaining four species(P<0.01).This difference in transcript levels might be due to the different pathogenesis of Mo in these breeds of sheep and goats.The abovefindings suggested that the susceptibility of different breeds of sheep and goats in Guizhou to Mo was related to the transcriptional levels of TLRs MyD88 pathway factors,providing basic research information for studying the pathogenic mechanisms of Mo in sheep and goats.
