摘要 PARP10是ADP-核糖基转移酶家族PARPs的一员,大量分布于细胞质和细胞核中,在细胞免疫应答、转录调控、DNA损伤修复、周期调节等生理过程中起着重要作用。PARP10可通过影响多种信号通路来促进肿瘤细胞的增殖与侵袭,已被发现参与了包括前列腺癌、乳腺癌、胃癌、肝癌等恶性肿瘤的发生过程。本研究通过生物信息学分析以及生化实验证明PARP10在结肠癌细胞的发生发展中也发挥重要作用。敲低PARP10的表达可以显著抑制肿瘤细胞增殖并促进其凋亡。通过筛选,我们发现香豆素(Coumarin)这一天然生物活性化合物可以抑制肿瘤细胞的增殖,并初步证明香豆素通过抑制结肠癌细胞中PARP10蛋白表达而降低结肠癌细胞的增殖能力。本研究为深入理解PARP10在结肠癌中作用以及相关的药物研发提供了新的思路。 PARP10 is a member of the ADP-ribosyltransferase family of PARPs and is widely distributed in both the cytoplasm and nucleus of cells.It plays an important role in cell immune response,transcriptional regulation,DNA damage repair,cell cycle regulation,and tumor proliferation.PARP10 promotes tumor cell proliferation and invasion by affecting various signaling pathways and has been found to be involved in the occurrence of malignant tumors such as prostate cancer,breast cancer,gastric cancer,and liver cancer.In this study,we determined that PARP10 also plays an important role in the proliferation of colorectal cancer cells through bioinformatics analysis and biochemical experiments.Inhibiting the expression of PARP10 can significantly inhibit tumor cell proliferation and promote apoptosis.Recently,it has been found that coumarin,a natural bioactive compound,can inhibit the proliferation of tumor cells,but its mechanism of anti-tumor action is unclear.In this study,we found that coumarin can target and decrease the expression of PARP10 protein in colon cancer cells,thereby inhibiting tumor growth.Therefore,this study provides new ideas and directions for colon cancer drug development and tumor treatment.
