大米免疫活性肽在小鼠体内的作用机制 被引量:2

A preliminary study on the effect mechanism of a rice immune peptide in mice

摘要 目的:证明前期筛选出的肽GBP1(NSVFRALPVDVVANAYR)在小鼠体内的免疫调节活性。方法:通过动物试验探讨GBP1肽的免疫调节作用机制;利用RNA-Seq技术对GBP1肽处理前后的小鼠脾脏组织进行转录组测序分析,阐释GBP1肽免疫调节作用的分子机制。结果:GBP1肽处理组小鼠脾细胞增殖数趋于正常值水平,肝脏器系数显著降低,血清中IgG含量稳定;终质量分数为25 mg/kg的GBP1肽能够在一定程度上抑制小鼠血清中IL-2、IFN-γ以及TNF-α的分泌;通过转录组分析,从脾脏组织中获得780个差异基因,该基因在机体内涉及与免疫相关的p53信号通路、铁死亡、补体和凝血级联等通路密切相关。结论:GBP1肽可以影响细胞因子的表达,介导信号转导通路,行使免疫调节功能。 Objective:This study aimed to demonstrate the potential immunomodulatory activity in mice of rice immunopeptide GBP1(NSVFRALPVDVVANAYR)screened previously.Methods:Animal experiments were conducted to investigate the immunomodulatory mechanism of GBP1 peptide.RNA-Seq was used to sequence the transcriptome of mouse spleen tissues before and after GBP1 peptide treatment,and the molecular mechanism of GBP1 peptide immunomodulatory effect was explained.Results:In the GBP1 peptide treatment group,the number of spleen cells tended to be normal,the liver organ coefficient decreased significantly,and the serum IgG content was stable.GBP1 peptide can affect the expression of cytokines,mediate signal transduction pathways,and exercise immune regulation function,and inhibit the secretion of IL-2,IFN-γand TNF-αin serum of mice.Through transcriptome analysis,780 differentially expressed genes were obtained from spleen tissue,which were closely related to immune-related p53 signaling pathway,ferroptosis,complement and coagulation cascade in the body.Conclusion:GBP1 peptide can affect the expression of cytokines,mediate signal transduction pathways,and exercise immune regulation function.
