绿脓菌素抑制缺铁环境中铜绿假单胞菌的生长代谢

摘要 为了探究铁离子对铜绿假单胞菌(PA)的影响,在缺铁及加铁条件下探究PA绿脓杆菌(PAO1)的生长状态,结果发现缺铁离子限制PAO1的生长.利用蛋白质组学质谱技术检测PAO1在不同铁离子条件下的蛋白表达差异,结果表明与合成绿脓菌素相关的吩嗪类蛋白在缺铁条件下表达量发生显著上调;定量检测结果验证PAO1在缺铁环境中产生绿脓菌素的量相较于加铁环境显著增加;通过检测外源加入的绿脓菌素对PAO1在缺铁及加铁条件下生长状态的影响,表明绿脓菌素会导致PAO1在生长初期生长缓慢且在生长中期菌体密度显著下降. PA生长状态的变化可能是由于缺铁条件下PAO1产生的绿脓菌素对其自身的杀伤作用所致. To investigate the effect of iron ion concentration on Pseudomonas aeruginosa,the growth state of P.aeruginosa reference strain(PAO1)was first explored under the conditions of iron deficiency and iron-rich,and the results showed that iron deficiency restricted the growth of PAO1.In order to explore this reason,the differentially-expressed proteins of P.aeruginosa PAO1 under different iron ion conditions were detected using proteomic mass spectrometry,and the data indicated that the expression of phenazine proteins related to the synthesis of pyocyanin was significantly upregulated under iron deficiency conditions.The quantitative detection results of pyocyanin further verified that the amount of pyocyanin produced by P.aeruginosa PAO1 in the iron-deficient environment was significantly increased compared with that in an iron-rich environment.By detecting the effect of exogenous addition of pyocyanin on the growth state of P.aeruginosa PAO1 under iro-deficient and iron-rich conditons,the data showed that pyocyanin could lead to a slow growth of P.aeruginosa PAO1 in the early stage of growth and a significant decrease in bacterial density in the middle stage.The results showed that the change in the growth state of P.aeruginosa might be due to the killing effect of pyocyanin produced by P.aeruginosa PAO1 under iron-deficient conditions.
