群体感应分子3OC12HSL对小鼠生长、血液和肠道的影响

Effects of Quorum Sensing Molecule N-3-Oxododecanoyl-L-Homoserine Lactone(3OC12HSL)on Growth,Blood and Gut of Mice

摘要 本研究旨在探讨群体感应分子3OC12HSL对小鼠生长、炎症因子及肠粘膜屏障的影响。采用高剂量盐酸林可霉素灌胃方法建立小鼠腹泻模型,对正常小鼠和腹泻模型小鼠分别灌胃生理盐水、二甲基亚砜(DMSO)、溶于DMSO的3OC12HSL溶液。结果显示,灌胃3OC12HSL降低正常小鼠和腹泻小鼠的增重,提高血清中D-乳酸、TNF-α含量,降低大肠长度及空肠和回肠绒毛长度(P<0.05);腹泻提高血清中IL-1、IL-8、TNF-α含量,降低空肠及回肠绒毛长度(P<0.05);灌胃3OC12HSL和腹泻对回肠绒隐比有协同降低趋势,但差异不显著(P>0.05)。综上,3OC12HSL不利于小鼠的生长,腹泻可能加重3OC12HSL的影响。 This study aimed to investigate the effects of quorum sensing molecule N-3-oxododecanoyl-L-homoserine lactone(3OC12HSL)on growth,inflammatory factors and intestinal mucosal barrier of mice.The mice diarrhea model was established by intragastric administration of high dose lincomycin hydrochloride solution.Normal and diarrhea mice were administrated intragastrically with normal saline,dimethyl sulfoxide(DMSO)or 3OC12HSL dissolved in DMSO,respectively.The results showed that intragastric administration of 3OC12HSL decreased weight gain of normal and diarrhea mice,increased serum D-lactate and TNF-α con-centrations,and decreased the length of large intestine and the villus length of jejunum and ileum(P<0.05).Diarrhea increased IL-1,IL-8 and TNF-α concentrations in serum,decreased the villus length of jejunum and ileum(P<0.05).3OC12HSL and diarrhea showed synergistically decreasing trend on the ratio of ileal villus length to crypt depth(P>0.05).In summary,3OC12HSL was not benefit to mice growth,and the influence might be exacerbated by diarrhea.
