高产多糖大球盖菇的ARTP诱变筛选及其菌丝培养条件优化

Screening of Stropharia rugosoannulata Strains with High Yield Polysaccharide by ARTP Mutagenesis and Optimization of Mycelial Culture Conditions

摘要 为获得优良的大球盖菇菌株,采用ARTP诱变处理大球盖菇孢子,通过检测诱变后的菌株菌丝生长势、拮抗作用及多糖含量,筛选菌丝生长快、多糖含量高的突变菌株;并对突变菌株菌丝多糖结构特征、红外光谱及菌丝液体培养条件进行分析。结果表明,在42株诱变菌株中,筛选得到5株生长势较好、遗传稳定,且与出发菌株有拮抗作用的突变菌株,其中菌株D1和D4的菌丝具有较高的生物量和多糖含量。D1菌株菌丝中的多糖颗粒均一、规则,粒径约为0.05μm,具有一般多糖的红外光谱特征;该菌株适宜的液体培养条件为孢子接种量8%、转速115 r·min-1、温度24℃、培养时间4 d,在此条件下菌丝生物量为56.82 mg。 In order to obtain excellent strains of Stropharia rugosoannulata,atmospheric and room temperature plasma mutagenesis was used to treat S.rugosoannulata spores.By exploring the growth vigor,antagonistic effects,and polysaccharide content of the mutated mycelium,the mutant strains with fast mycelial growth and high polysaccharide content were screened,and the mycelial polysaccharide structural feature and infrared spectroscopy,and mycelium liquid culture conditions of the mutant strain were investigated.The results showed that among the 42 screened mutagenic strains,5 mutant strains with good growth potential,genetic stability and antagonism to the original strain were obtained.Among them,the mycelium of D1 and D4 strains had higher biomass and polysaccharide content,the mycelium polysaccharide particles of D1 strain were uniform and regular,and the particle size was about 0.05μm,which had the infrared spectrum characteristics of general polysaccharides.The suitable liquid culture conditions for D1 strain mycelium are spore inoculation rate of 8%,rotation speed of 115 r·min-1,culture temperature of 24℃,and culture time of 4 days,the biomass of mycelium under these conditions was 56.82 mg.
