柳氮磺胺吡啶联合脆弱拟杆菌治疗对溃疡性结肠炎小鼠脾脏中Treg比例的影响

The effect of sulfasalazine combined with Bacteroides fragilis treatment on the proportion of Treg in the spleen of mice with ulcerative colitis

目的:探究柳氮磺胺吡啶联合脆弱拟杆菌治疗对溃疡性结肠炎小鼠脾脏中Treg比例的影响。方法:研究时间为2020年1月1日至2022年12月31日,选取32只BALB/c小鼠作为本研究的实验对象,按照不同处理方法分为对照组、模型组、单纯治疗组、联合组(柳氮磺胺吡啶联合脆弱拟杆菌),各8只。对照组不做处理,模型组、单纯治疗组、联合组均使用葡萄糖硫酸钠盐制作慢性溃疡性结肠炎小鼠。模型组实施建模,单纯治疗组在模型组的基础上每天给予柳氮磺胺吡啶治疗,联合组在建模的基础上每天同时给予柳氮磺胺吡啶、脆弱拟杆菌治疗。治疗7周后处死小鼠,取出小鼠的结肠组织进行检测,并对比4组疾病活动指数(DAI)、结肠黏膜损伤指数(CMDI)、结肠组织A20蛋白表达量及脾脏组织Treg比例。结果:与对照组相比,模型组DAI评分、CMDI评分均升高,且A20蛋白相对表达量与Treg比例均降低;与模型组对比,单纯治疗组DAI评分、CMDI评分均降低,且A20蛋白相对表达量与Treg比例均升高;与单纯治疗组对比,联合组DAI、CMDI评分显著降低,且A20蛋白相对表达量与Treg比例均升高。结论:在溃疡性结肠炎小鼠的研究中发现,使用柳氮磺胺吡啶、脆弱拟杆菌治疗具有较高的效果,可以有效改善小鼠的脾脏组织中Treg比例。

Objective:To investigate the effect of sulfasalazine combined with Bacteroides fragilis treatment on the proportion of Treg in the spleen of mice with ulcerative colitis.Method:The study period was from January 1,2020 to December 31,2022.32 BALB/c mice were selected as the experimental subjects of this study.They were divided into a control group,a model group,a simple treatment group,and a combination group(sulfasalazine combined with Bacteroides fragilis)according to different treatment methods,with 8 mice in each group.The control group was not treated,while the model group,simple treatment group,and combination group were all treated with sodium glucose sulfate to produce chronic ulcerative colitis mice.The model group implemented modeling,while the simple treatment group received daily treatment with sulfasalazine on the basis of the model group,while the combination group received daily treatment with sulfasalazine and Vibrio fragilis on the basis of modeling.After 7 weeks of treatment,the mice were euthanized and their colon tissue was taken out for testing.The disease activity index(DAI),colon mucosal injury index(CMDI),A20 protein expression in colon tissue,and Treg ratio in spleen tissue were compared among the four groups.Result:Compared with the control group,the DAI score and CMDI score of the model group increased,and the relative expression level of A20 protein and the proportion of Treg decreased;Compared with the model group,the DAI score and CMDI score of the simple treatment group decreased,and the relative expression level of A20 protein and the proportion of Treg increased;Compared with the simple treatment group,the combination group showed a significant decrease in DAI and CMDI scores,and the relative expression level of A20 protein and Treg ratio increased.Conclusion:In the study of ulcerative colitis mice,it was found that treatment with sulfasalazine and Bacteroides fragilis had a high effect and could effectively improve the Treg ratio in the spleen tissue of mice.
