不同浓度碘对甲状腺癌细胞增殖的影响

摘要 目的探讨不同浓度碘对甲状腺癌细胞增殖的影响。方法采用不同碘浓度培养BCPAP细胞,并分为对照组(不含碘)、高碘浓度组(含碘1.0×10^(-3)mol/L)和适量碘浓度组(含碘1.0×10^(-6)mol/L),检测BCPAP细胞增殖能力;采用高通量测序方法检测BCPAP细胞RNA,比较不同碘浓度对BCPAP细胞增殖的影响及细胞差异基因的表达情况。结果高碘浓度组BCPAP细胞增殖能力低于对照组和适量碘浓度组,而适量碘浓度组高于对照组,差异有统计学意义(P<0.05)。高碘浓度组与对照组细胞间共有522个差异表达基因,其中上调基因266个,下调基因256个;适量碘浓度组与对照组细胞间共有552个差异表达基因,其中上调基因10个,下调基因542个。结论高浓度碘抑制甲状腺癌细胞增殖起,适量浓度碘可促进甲状腺癌细胞增殖,高碘浓度组和适量碘浓度组皆有差异表达基因。 Objective To explore the effect of different concentrations of iodine on the proliferation of thyroid cancer cells.Methods BCPAP cells were cultured with different iodine concentrations and divided into the control group(without iodine),the high iodine concentration group(1.0×10^(-3) mol/L)and the appropriate iodine concentration group(1.0×10^(-6) mol/L),and the proliferation ability of BCPAP cells was detected;BCPAP cell RNA was detected by high-throughput sequencing method,and the effects of different iodine concentrations on BCPAP cell proliferation and the ex-pression of differential genes were compared.Results The proliferation capacity of BCPAP cells in the high iodine concentration group was lower than that in the control group and the appropriate iodine concentration group,while the appropriate iodine concentration group was higher than the con-trol group,the differences were statistically significant(P<0.05).There were 522 differentially expressed genes between the high iodine concentration group and the control group,including 266 up-regulated genes and 256 down-regulated genes.There were 552 differentially expressed genes between the appropriate iodine concentration group and the control group,including 10 up-regulated genes and 542 down-regulated genes.Conclusion High iodine concentration inhibits the proliferation of thyroid cancer cells,while appropriate iodine concentration promotes the proliferation of thyroid can-cer cells,there were differentially expressed genes in between the high iodine concentration group and the appropriate iodine concentration group.
