基于网络药理学探讨桑白皮-夏枯草治疗原发性高血压的作用机制

摘要 目的 此研究旨在通过网络药理学,探究桑白皮-夏枯草对原发性高血压的有效靶标及作用机制。方法 从中药系统药理学数据库收集桑白皮-夏枯草化学成分及对应靶标,进行精确筛选。运用全球蛋白质资源数据库将靶标转化为人类基因简称。通过研究基因卡片数据库中原发性高血压靶标,找到其与药对靶标的交集,使用生物分子功能注释系统构建蛋白互作网络,上传Cytoscape确定核心靶标,后对其开展基因本体功能研究和基于京都基因与基因组百科全书通路富集分析。结果 共发现39个有效成分,41个核心靶标,454个生物过程及143条通路。结论 桑白皮-夏枯草对原发性高血压有多靶标、多信号通路协同作用,为进一步研究和指导临床用药提供了理论依据。 Objective Through the network pharmacology to explore targets and mechanism of Cortex Mori-Spica Prunellae in the treatment of essential hypertension.Methods The Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform was utilized to discover and filter the effective constituents and targets of the drugs.The global Protein Resource database was employed to convert each target site into its corresponding human gene abbreviation.The disease targets of essential hypertension were retrieved through Genecards database.The biomolecular function annotation system database was used to upload the intersection of drugs and disease targets,which then formed a PPI protein interaction network.Uploading the network to Cytoscape to identify the core targets,which were analyzed by gene ontology function analysis and pathway enrichment analysis based on the Kyoto Encyclopedia of Gene and Genome.Results In total,39 active components,41 core targets,454 biological processes and 143 pathways were found.Conclusion Cortex Mori-Spica Prunellae demonstrate a synergistic effect on the treatment of essential hypertension through multiple targets and signal pathways,thus providing a basis for further exploration and direction of clinical medication.
