原花青素添加对鸡精液冷冻效果的影响

摘要 为探究抗氧化剂原花青素(Procyanidin,PC)在鸡精液冷冻中的应用效果,在10%卵黄上清稀释液中添加不同质量浓度(10μg∕mL、20μg∕mL、30μg∕mL、40μg∕mL、60μg∕mL)的PC,检测冻后鸡精子的质量指标和抗氧化指标。结果表明:在卵黄上清稀释液中添加10—30μg∕mL PC,冻后鸡精子的质量指标(存活率、活力、质膜完整性、顶体完整性和线粒体活性)与对照之间均无显著差异。当PC添加质量浓度为40—60μg∕mL时,冻后鸡精子的各项质量指标均显著下降(P<0.05)。在抗氧化指标方面,PC添加可提高冻后鸡精子的抗氧化能力,其中添加30μg∕mL PC的冻后鸡精子抗氧化能力最高,冻后鸡精子的MDA含量和ROS含量最低,分别为5.48 nmol∕L和342.00 U∕mL;SOD活性和GSH-PX活性最高,分别为137 U∕mL和90.85 U∕L,且与其他各组差异显著。当PC添加质量浓度为40—60μg∕mL时,冻后鸡精子的抗氧化能力显著下降,表现为对精子的毒副损伤。可见,在鸡精液冷冻过程中添加10—30μg∕mL的PC虽不能显著提高冻后鸡精子的质量,但却能显著提高其抗氧化能力。 In order to explore the application effects of antioxidant procyanidin(PC)in rooster semen freezing,different concentrations of PC(10μg∕mL,20μg∕mL,30μg∕mL,40μg∕mL,60μg∕mL)were added to 10%yolk supernatant dilution,the quality indexes and antioxidant indexes of frozen sperm were detected.The results showed that compared to the control,there was no significant difference in the quality indexes(survival rate,viability,membrane integrity,acrosome integrity,and mitochondrial activity)of frozen rooster sperm when 10—30μg∕mL PC was added to the yolk supernatant dilution.When adding PC with 40—60μg∕mL,all quality indexes of frozen chicken sperm significantly decreased(P<0.05).In terms of antioxidant indexes,the addition of PC could improve the antioxidant capacity of frozen rooster sperm,among which the antioxidant capacity of frozen chicken sperm with 30μg∕mL PC addition was the highest.The MDA content and ROS content of frozen rooster sperm were the lowest,at 5.48 nmol∕L and 342.00 U∕mL,respectively;SOD activity and GSH-PX activity were the highest,at 137 U∕mL and 90.85 U∕L,respectively,with significant differences compared to other groups.When the concentration of PC added was 40—60μg∕mL,the antioxidant capacity of frozen rooster sperm significantly decreased,showing toxic side effects on sperm.It could be seen that adding 10—30μg∕mL PC during the freezing process of rooster semen may not significantly improve the quality of frozen rooster sperm,but it can significantly enhance its antioxidant capacity.
