摘要
目的:检测LINC01535基因在上皮性卵巢癌(epithelial ovarian cancer, EOC)组织及细胞株中的表达情况及其与患者临床病理和预后的关系,探讨LINC01535对上皮性卵巢癌细胞CAOV3增殖、侵袭及EMT的影响。方法:基于GEPIA2和Kaplan-Meier Plotter数据库分析LINC01535在上皮性卵巢癌组织表达情况及对卵巢癌患者生存期的影响,应用实时荧光定量qRT-PCR(qRT-PCR)的方法检测上皮性卵巢癌组织和正常组织以及三种上皮性卵巢癌细胞株(SKOV3、A2780、和CAOV3)中LINC01535基因的表达水平,分析LINC01535基因表达与临床病理参数及预后的关系;应用siRNA瞬时转染上皮性卵巢癌细胞CAOV3干扰LINC01535基因的表达后,MTS和Transwell小室迁移和侵袭实验检测LINC01535基因异常表达对上皮性卵巢癌细胞CAOV3增殖、迁移和侵袭的影响。qRT-PCR法和Western blot法检测LINC01535对于EMT过程相关标志物(E-cadherin、N-cadherin及Vimentin)表达水平的影响。结果:应用GEPIA2和Kaplan-Meier Plotter数据库发现LINC01535在上皮性卵巢癌肿瘤组织中的表达显著高于正常卵巢组织(P<0.01),LINC01535高表达组的总体生存率显著低于LINC01535低表达组(P<0.01);LINC01535基因在89例上皮性卵巢癌组织中的表达明显高于正常组织(P<0.01),并与大网膜转移、淋巴结转移、FIGO分期及生存期相关(P<0.01或P<0.05),敲低LINC01535基因后可以抑制上皮性卵巢癌细胞系CAOV3的体外增殖、迁移、侵袭能力及EMT进程(P<0.01或P<0.05)。结论:LINC01535基因在上皮性卵巢癌中可能发挥癌基因的作用,其高表达可能促进了上皮性卵巢癌细胞的增殖、侵袭及EMT进程,并有望成为上皮性卵巢癌患者预后评估的候选分子标记物。
Objective:To detect the expression of LINC01535 gene in epithelial ovarian cancer(EOC)tissues and cell lines,as well as its relationship with clinical pathology and prognosis of patients.To explore the impact of LINC01535 on the proliferation,invasion,and EMT of epithelial ovarian cancer cell line CAOV3.Methods:Based on the GEPIA2 and Kaplan-Meier Plotter databases,the expression of LINC01535 in epithelial ovarian cancer tissue and its impact on survival of ovarian cancer patients were analyzed.The real-time fluorescence quantitative qRT-PCR method was used to detect the expression level of LINC01535 gene in epithelial ovarian cancer tissue,normal tissue,and three epithelial ovarian cancer cell lines(SKOV3,A2780,and CAOV3).The relationship between LINC01535 gene expression and clinical pathological parameters and prognosis was analyzed.After using siRNA transient transfection to interfere with the expression of LINC01535 gene in epithelial ovarian cancer cells CAOV3,MTS and Transwell cell experiments were performed to detect the impact of abnormal expression of LINC01535 gene on the proliferation,migration,and invasion of epithelial ovarian cancer cell CAOV3.The effect of LINC01535 on the expression levels of EMT process related biomarkers(E-cadherin,N-cadherin,and Vimentin)was detected using qRT-PCR.Results:Using GEPIA2 and Kaplan-Meier Plotter databases,it was found that the expression of LINC01535 in epithelial ovarian cancer tumor tissue was significantly higher than that in normal tissue(P<0.05),and the overall survival rate of the high expression group of LINC01535 was significantly lower than that of the low expression group of LINC01535(P<0.01).The expression of LINC01535 gene in 89 cases of epithelial ovarian cancer tissue was significantly higher than that of normal tissue(P<0.01),and was associated with greater omentum metastasis,lymph node metastasis,FIGO staging,and survival(P<0.01 or P<0.05).Knocking down LINC01535 gene can inhibit the in vitro proliferation,migration,invasion ability and EMT process of epithelial ovarian cancer cell line CAOV3(P<0.01 or P<0.05).Conclusion:LINC01535 gene may play a role as an oncogene in epithelial ovarian cancer.Its high expression may promote the proliferation,invasion,and EMT process of epithelial ovarian cancer cells,and may become a candidate molecular marker for prognostic evaluation of epithelial ovarian cancer patients.
