微小RNA-132在胰腺癌患者外周血中的表达及其临床意义

摘要 目的研究微小RNA-132在胰腺癌患者外周血中的表达和临床意义。方法选择83例胰腺癌患者作为观察组,另外选择接受治疗的50例胰腺炎患者和50例健康体检者作为对照组Ⅰ组和对照Ⅱ组。所有研究对象均采用Trizol法提取细胞当中的微小RNA-132,并通过反转录试剂盒反转录成cDNA,根据实时荧光定量聚合酶链反应(RT-qPCR)试剂盒说明书配制反应体系进行扩增,计算miR-132的相对表达水平。对比三组微小RNA-132相对表达水平,分析微小RNA-132相对表达对胰腺癌的诊断价值。结果①三组微小RNA-132相对表达水平对比,差异有统计学意义(P<0.05);观察组外周血中的微小RNA-132相对表达水平(2.87±0.60)明显高于对照组Ⅰ组的(2.50±0.52)和对照Ⅱ组的(2.02±0.42),对照Ⅰ组高于对照组Ⅱ组,差异有统计学意义(P<0.05)。②受试者工作特征曲线(ROC)分析可得,微小RNA-132诊断胰腺癌的最佳临界点为2.79,外周血的微小RNA-132相对表达水平达到2.79,其诊断胰腺癌的曲线下面积(AUC)为0.907,95%CI=(0.864,0.949),敏感度为79.50%,特异度为90.00%,P<0.05。结论胰腺癌患者外周血中的微小RNA-132有明显上调趋势,可将微小RNA-132当做是对胰腺癌患者进行潜在诊断的主要标记物。 Objective To study the expression of microRNA-132 in peripheral blood of patients with pancreatic cancer and its clinical significance.Methods 83 patients with pancreatic cancer were selected as the observation group,and 50 patients with pancreatitis and 50 healthy subjects were selected as the control groupⅠand control groupⅡ.All study subjects were extracted microRNA-132 from cells by Trizol method and reverse-transcribed into cDNA by reverse transcription kit.A reaction system was prepared according to the instructions of the real-time quantitative polymerase chain reaction(RT-qPCR)kit for amplification,and the relative expression level of microRNA-132 was calculated.The relative expression levels of microRNA-132 in the three groups were compared to analyze the diagnostic value of relative expression of micro RNA-132 for pancreatic cancer.Results(i)The relative expression level of microRNA-132 was compared among the three groups,and the difference was statistically significant(P<0.05).The relative expression level of microRNA-132 in peripheral blood of the observation group was(2.87±0.60),which was significantly higher than(2.50±0.52)of the control groupⅠand(2.02±0.42)of the control groupⅡ;the control groupⅠwas higher than the control groupⅡ;the difference was statistically significant(P<0.05).(ii)Receiver operating characteristic(ROC)curve showed that the optimal threshold for the diagnosis of pancreatic cancer by micro RNA-132 was 2.79;the relative expression level of microRNA-132 in peripheral blood reached 2.79,and its area under the curve(AUC)for the diagnosis of pancreatic cancer was 0.907,95%CI=(0.864,0.949),with a sensitivity of 79.50%and a specificity of 90.00%,P<0.05.Conclusion There is a significant trend of upregulation of microRNA-132 in the peripheral blood of patients with pancreatic cancer,and microRNA-132 can be used as a primary marker for the potential diagnosis of patients with pancreatic cancer.
