摘要
该研究旨在探究酶解鲍鱼肽的分子特性及其抗氧化、调节免疫的功能。通过酶解获得鲍鱼多肽,利用液相色谱、氨基酸分析仪分别测定了鲍鱼肽分子质量和氨基酸含量,并在小鼠体内研究了鲍鱼肽抗氧化和调节免疫的功能,均设置低(0.25 g/kg BW)、中(0.5 g/kg BW)、高(1.0 g/kg BW)3个剂量组,以及对应的对照组。研究结果显示鲍鱼肽的分子质量低于6500 Da占95%,氨基酸含量为72.54%。体内实验结果显示,抗氧化实验中,3个剂量组小鼠血清中的丙二醛和蛋白质羰基含量显著低于模型组,超氧化歧化酶和还原性谷胱甘肽含量显著高于模型组。在调节免疫实验中,与模型组相比,中、高剂量组小鼠白细胞数显著高于模型组,各组细胞免疫功能均无显著差异,体液免疫中,高剂量组小鼠脾细胞中半数溶血值和溶血空斑数量均显著提高,且中、高剂量组的NK细胞活性显著提高。综上所述,鲍鱼肽的分子质量小且氨基酸含量高,具有抗氧化和调节免疫的功能。
The aim of this study was to investigate the molecular properties of zymolytic abalone peptide and its functions of antioxidant and immunological.After enzymolysis,the molecular weight and amino acid content of the abalone peptide were determined by liquid chromatography and an amino acid analyzer,and the antioxidant and immunological functions of the abalone peptide were studied in mice.Three dose groups and the corresponding control group were set up as low(0.25 g/kg BW),medium(0.5 g/kg BW),and high(1.0 g/kg BW).Results showed that the molecular weight of abalone peptide was less than 6500 Da accounting for 95%,and the amino acid content was 72.54%.In vivo experiments showed that the contents of malondialdehyde(MDA)and protein carbonyl(PCO)in the serum of mice in the three dose groups were significantly lower than those in the model group,and the contents of superoxide dismutase(SOD)and reducing glutathione(GSH)were significantly higher than those in the model group.In the regulated immunity experiment,compared with the model group,the leukocyte count of mice in the medium-dose and high-dose groups was significantly higher than that in the model group,and there was no significant difference in cellular immune function among all groups.The half hemolysis value and the number of hemolytic plaques in the spleen cells of mice in the medium-and the high-dose group were significantly increased,and the activity of NK cells was also significantly increased.In conclusion,the tested abalone peptide has a small molecular weight and high amino acid content and has antioxidant and immune regulatory functions.
