瓜石汤联合当归芍药散对多囊卵巢综合征合并胰岛素抵抗小鼠的影响

摘要 目的:探讨瓜石汤联合当归芍药散对多囊卵巢综合征联合胰岛素抵抗(PCOS-IR)小鼠模型性激素和内膜炎症介质的影响。方法:用来曲唑和高脂饮食构建PCOS-IR模型小鼠,观察小鼠动情周期变化及检测小鼠空腹血糖、空腹胰岛素评价模型成功是否。将小鼠随机分为对照组、模型组、中药组,中药组造模成功后给予瓜石汤联合当归芍药散中药方灌胃,对照组及模型组给予等体积纯净水。干预21 d后用酶联免疫吸附试验(ELISA)法测定血清卵泡刺激素(FSH)、促黄体生成素(LH)、总睾酮(T)水平,检测内膜组织肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)的水平。结果:造模后对照组小鼠动情周期正常,模型组小鼠动情周期紊乱,多数位于动情间期;对照组小鼠胰岛素抵抗指数(HOMA-IR)均小于2.69,而模型组均大于2.69。给药21 d后,中药组小鼠血清LH、T、LH/FSH较模型组低(P<0.05),内膜组织TNF-α、IL-1β、IL-6 mRNA和蛋白表达水平较模型组低(P<0.05)。结论:瓜石汤联合当归芍药散能改善PCOS-IR小鼠血清激素水平,降低内膜炎症介质水平,改善内膜炎症。 Objective:To explore the effects of Guashi Decoction combined with Danggui Shaoyao Powder on sex hormones and endometrial inflammatory factors in mice models with polycystic ovary syndrome combined with insulin resistance(PCOS-IR).Methods:PCOS-IR mice models were constructed with letrozole and a high-fat diet,and the success of the model was evaluated by observing changes in the estrous cycle and measuring fasting blood glucose and fasting insulin in mice.The mice were randomly divided into a control group,model group,and traditional Chinese medicine(TCM)group,and the TCM was given Guashi Decoction combined with Danggui Shaoyao Powder by gavage after successful modeling,while the control group and model group were given an equal volume of pure water.Serum follicle-stimulating hormone(FSH),luteinizing hormone(LH),and total testosterone(T)levels were measured after 21 days of intervention by enzyme-linked immunosorbent assay(ELISA)method,and the levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),and interleukin-6(IL-6)in endothelial tissue were measured.Results:After modeling,the mice in the control group had a normal estrous cycle,while the mice in the model group had a disturbed estrous cycle,mostly in the interestrous phase.The homeostasis model assessment-insulin resistance(HOMA-IR)of the control mice was less than 2.69,while that in the model group was greater than 2.69.After 21 days of the administration,the serum LH,T,and LH/FSH of mice in the TCM group were lower than those in the model group(P<0.05),and the mRNA and protein expression levels of TNF-α,IL-1β,and IL-6 in endothelial tissue were lower than those in the model group(P<0.05).Conclusion:Guashi Decoction combined with Danggui Shaoyao Powder can improve the serum hormone level,reduce the level of endothelial inflammatory factors,and improve the endothelial inflammatory status in PCOS-IR mice.
