节律基因PER3基于胰腺癌干细胞调控的胰腺癌转移抑制

摘要 胰腺癌是一种极具破坏性的消化系统癌症.PER3是昼夜节律基因家族的主要成员,世界卫生组织将其列为潜在的致癌物.有报道显示PER3的异常表达与结直肠癌的发生发展有关,为了解其是否参与胰腺癌调控,使用小鼠胰腺癌细胞系Pan02构建PER3低表达细胞系,利用MTT法证明敲低PER3会抑制细胞增殖.通过Transwell实验证明敲低PER3能够抑制细胞迁移.细胞成球实验证明敲低PER3降低了胰腺癌干细胞的干性,并且抑制了干性标记物mRNA的表达.以上结果表明节律基因PER3可以通过抑制胰腺癌干细胞的干性进而抑制胰腺癌转移. Pancreatic cancer is a destructive cancer of the digestive system.PER3 is a major member of the circadian gene family,which is listed as a potential carcinogen by the World Health Organization.It has been reported that the abnormal expression of PER3 is related to the occurrence and development of colorectal cancer.To investigate its role in pancreatic cancer regulation,a mouse pancreatic cancer cell line Pan02 is used to construct a cell line with low expression of PER3.MTT assay is used to prove that knockdown PER3 can inhibit cell proliferation.Transwell assay is used to prove that knockdown PER3 inhibits cell migration.Cell ball formation experiments show that knockdown PER3 decreases the stemness of pancreatic cancer stem cells and inhibits the expression of stemness marker mRNA.These results suggest that the circadian gene PER3 inhibits the metastasis of pancreatic cancer by inhibiting the stemness of pancreatic cancer stem cells.
