摘要
重组病毒载体是当前病毒基因工程研究的热点之一。羊口疮(orf),也称羊传染性脓疱皮炎,是由羊口疮病毒(orf virus,ORFV)引起的急性、接触性、嗜上皮性人畜共患传染病。ORFV为痘病毒科(Poxviridae)、脊索动物痘病毒亚科(Chordopoxrinae)的副痘病毒属(Parapoxvirus)成员,线性双股DNA病毒,病毒基因组较大,改造后可容纳较大的外源基因,适合开发为活病毒载体。本文综述了orf重组活病毒载体的优势、构建策略及其应用研究的最新进展,为ORFV研究、重组活病毒开发应用提供参考。
Recombinant virus vector has become one of the current hotspots in viral genetic engineering.Orf,also known as contagious pustular dermatitis,is an acute,contact,epitheliophilic zoonoses that is caused by orf virus(ORFV),the type species of the genus Parapoxvirus within the poxvirus subfamily Chordopoxvirinae.ORFV is linear double-stranded DNA(dsDNA)virus with a large viral genome that can accommodate large exogenous genes after transformation.It is suitable for developing as a live viral vector.In this paper,the advantages,vector modification strategies,and application of recombinant live viral vector were reviewed,which can provide reference for the research of ORFV and the development and application of recombinant live virus.
