摘要
为了研究肽聚糖相关脂蛋白(peptidoglycan-associated lipoprotein,Pal)在肠炎沙门菌生物被膜形成和耐药中的作用,本研究构建了肠炎沙门菌的pal缺失株,对其生物被膜形成能力进行检测,同时检测Curli菌毛和纤维素形成,以及检测生物被膜形成相关基因表达水平。结果显示,与野生型菌株相比,pal基因缺失株生物被膜形成能力显著降低,Curli菌毛分泌量降低,但纤维素形成无显著降低,生物被膜相关基因表达水平显著降低。为进一步分析pal基因在肠炎沙门菌耐药性中的作用,本研究测试了多黏菌素B对该菌的最低抑菌浓度(MIC)。结果显示,pal基因缺失株对多黏菌素B的MIC降低了75%。综上所述,本研究表明pal基因影响肠炎沙门菌生物被膜形成和耐药性,研究结果为肠炎沙门菌Pal的功能研究奠定了基础。
In order to investigate the role of peptidoglycan-associated lipoprotein(Pal)in biofilm formation and drug resistance of Salmonella enterica,a pal-deficient strain of Salmonella enterica was constructed,and its ability to form biofilm,the formation of Curli hairs and fibrils and the expression levels of biofilm-related genes were examined.The results showed that compared with the wild-type strain,the pal-deficient strain had significantly lower biofilm formation ability,lower secretion of Curli hairs,but no significant reduction in cellulose formation,and significantly lower expression levels of biofilm-related genes.To further analyze the role of pal gene in Salmonella enterica drug resistance,the lowest inhibitory concentration of polymyxin B against this bacterium was tested.The results showed that the pal-deficient strain showed approximately 75%lower MIC against polymyxin B.In summary,this study showed that the pal gene affects Salmonella enterica biofilm formation and drug resistance,and the results lay the foundation for functional studies of Salmonella enterica Pal.
