基于糖化酶的布鲁氏菌抗体快速检测方法的建立与评估

摘要 目的:建立一种布鲁氏菌抗体相对定量检测的新方法,并对其性能进行评价。方法:验证液化条件;建立糖化酶与血糖仪反应值的标准曲线;制备酶标抗体及ELISA平板进行样本检测及反应体系干扰验证的试验。结果:糖化酶浓度与糖浓度成正比,糖化酶-布鲁氏菌脂多糖抗原耦联-1对阴阳性区分度更好,干扰验证试验发现临床样本中葡萄糖、HBsAg、HBsAb、HCV和BSA的水平,均对检测结果无影响。结论:通过构建基于葡萄糖检测的ELISA法,通过便捷式血糖仪,可以实现快速检测布鲁氏菌抗体,为临床诊断提供了新的手段。 Objective:To Establish a new method for relative quantitative detection of Brucella antibodies and evaluate its performance.Method:Verify liquefaction conditions;Establish the standard curve of the reaction value between glucoamylase and Glucose meter;Preparation of enzyme-linked antibodies and ELISA plates for sample detection and validation of reaction system interference.Results:The concentration of glucoamylase is in direct proportion to the concentration of sugar,and the glucoamylase Brucella Lipopolysaccharide antigen coupling-1 has better discrimination between yin and yang.The interference verification test found that the levels of glucose,HBsAg,HBsAb,HCV and BSA in clinical samples have no impact on the detection results.Conclusion:By constructing an ELISA based on glucose detection and using a convenient Glucose meter,we can quickly detect brucella antibodies,providing a new means for clinical diagnosis.
